This document discusses opportunities and challenges for using biomarkers to accelerate proof-of-concept studies in neurodegenerative diseases. It highlights initiatives like ADNI that have advanced biomarkers for disease state and progression. These biomarkers allow for earlier patient enrichment, more sensitive endpoints, and smaller clinical trials. Biomarkers can also provide insights into drug-target engagement and pharmacodynamics/toxicodynamics. Integrating biomarkers throughout development could enable faster entry to Phase 3 and higher chances of success for disease-modifying therapies to address the growing burden of neurodegenerative diseases.
This document summarizes several studies that evaluated the use of ocular ultrasound to measure optic nerve sheath diameter (ONSD) as a non-invasive method to detect raised intracranial pressure compared to CT scan findings. The studies reported sensitivities ranging from 74% to 100% and specificities ranging from 63% to 100% for using ONSD cut-off values of 5.0-5.9mm to diagnose raised ICP. However, limitations included small sample sizes, potential for selection bias, and lack of blinding in some studies. Larger, blinded studies are still needed to validate ONSD as a reliable alternative to CT scans.
This document discusses several controversies in Alzheimer's disease research and treatment approaches. It notes that β-amyloid deposits are often found diffusely in normal elderly individuals without cognitive impairment. Both amyloid and tau appear necessary for amyloid oligomers to induce neuronal changes and dementia. While recent drug trials failed, the document argues that targeting earlier mild disease stages and testing multiple candidates may lead to success in the future.

Presentation at the VGCt/VVGT Najaarscongres in Veldhoven, The Netherlands, at November 18, 2011, entiteled "The place and finetuning of (experiential) exposure as an intervention for borderline patients: Predictions from empirical studies"
1) The study examined overgeneral memory in borderline personality disorder patients and found relationships between overgeneral memory and depression severity and rumination that were consistent with findings in depressed patients.
2) It also found that the more an autobiographical memory cue was relevant or discrepant with the patient's ideal self, the more difficulty they had retrieving specific memories, as seen in depressed patients.
3) Depression severity appeared to play an important role in overgeneral memory in borderline personality disorder patients.
Test for HIV-associated cognitive impairment in IndiaKimberly Schafer
This document describes the development of a brief screening battery to detect HIV-associated neurocognitive impairment (HAND) in India. Researchers administered a comprehensive neuropsychological (NP) battery to 206 HIV-positive Indian patients. Statistical analysis identified that combinations of two tests - the Brief Visuospatial Memory Test-Revised for learning and either the Color Trails 1 test for processing speed, Grooved Pegboard test for motor skills, or Digit Symbol test for processing speed - achieved high sensitivity and specificity for detecting HAND. The study aims to develop a quick iPad-based screening tool to assess cognitive functioning in resource-limited settings like India.
JH Alzheimer's and Parkinson's disease meeting presentation Florence March 2013John Harrison
This presentation was made at the AD/PD meeting in Florence on 8th March 2013. In the presentation I discuss early patterns of cognitive dysfunction in patients with Parkinson's disease and Alzheimer's disease. I describe the tests traditionally used to measure cognitive impairment and propose the use of an assessment with the potential to be used successfully in both indications. I propose also that the same collection of measures can profitably be used in other CNS indications. I stress that selection is best made on the basis not of specific tests, but instead on the use of good tests that meet current best practice guidance for measuring cognitive change.

Il ruolo del pensiero desiderante lungo il continuum della dipendenza da alcool
Whole brain radiation therapy (WBRT) plays an important role in the treatment of brain metastases and claims of its obsolescence are exaggerated. While stereotactic radiosurgery (SRS) alone has advantages like reduced cognitive side effects, it also has significantly higher rates of local and distant brain recurrence requiring additional treatment. Studies comparing SRS alone to SRS with WBRT have shown WBRT improves intracranial control and in some cases survival, especially in patients with better prognosis and those with non-small cell lung cancer. New techniques like hippocampal-avoidance WBRT and memantine may further reduce cognitive toxicity when combined with WBRT, providing a more favorable risk-benefit profile

The document presents a method for automatically mapping terms from clinical encounter forms to concepts in SNOMED CT. It exploits the semantic structure of forms by analyzing the context and relationships between terms. A naive Bayes classifier is trained on semantic attributes derived from the form structure to determine the appropriate SNOMED CT semantic category for each term. Evaluation on 26 forms showed the hybrid approach of combining linguistic techniques and semantic structure outperformed a baseline, achieving a precision of 0.89 and recall of 0.76.
HPCs have difficulty with the large amounts of data generated from shotgun sequencing of biological samples. This document discusses three main computational problems with sequencing data: resequencing analysis to find variants, counting sequences to determine abundance, and de novo assembly without a reference. It proposes using lossy compression algorithms and streaming analysis to reduce data sizes and memory requirements while retaining essential information, enabling analysis on commodity hardware rather than expensive HPCs. The approaches developed in the author's lab have shown effectiveness in testing on real sequencing data sets.

MRI Workshop
Dr. Ben Turner held an MRI workshop in November 2018. He discussed the principles of magnetic resonance imaging, different MRI techniques like T2 imaging and flair images, and how MRI is beneficial but also has drawbacks for monitoring multiple sclerosis. MRI is most useful for research, diagnosis, therapeutic innovation for drug trials, and monitoring therapies. New diagnostic criteria for multiple sclerosis were also presented, focusing on dissemination of lesions in space and time with no better explanation. Factors like lesion number and location provide prognostic information about progression.

Overgeneral memory (OGM) refers to the tendency to recall categories of events from memory, rather than specific memories, even when it is explicitly instructed to recall specific events. Unlike depressed and traumatized patients, patients diagnosed with borderline personality disorder (BPD) inconsistently show OGM. Non-suicidal self-injury (NSSI) on the other hand, is very common in patients with BPD. Like OGM, NSSI is considered to be an affect-regulation mechanism. This study investigated how these strategies relate to each other. Based on earlier findings (Startup et al., 2001), we hypothesized that NSSI and OGM would be inversely associated. Fifty three BPD patients completed the Structured Clinical Interview for DSM-IV Disorders (Axis II as well as Axis I-mood modules), the Autobiographical Memory Test to assess OGM, and the Self-Injury Questionnaire - Treatment Related (SIQ-TR) to assess NNSI. We found that patients who engage in NSSI do not differ from patients who do not with respect to OGM. However, we found that participants who use more different NSSI methods showed less OGM, but this association disappeared when we controlled for age. We propose a balance-model of affect-regulation as one possible explanation for the relationship between these two affect-regulation behaviours, and discuss the clinical relevance of our findings.
The document summarizes a study on overgeneral memory in patients with borderline personality disorder (BPD). The study examined: (1) whether overgeneral memory (OGM) in BPD is associated with major depressive disorder (MDD) or post-traumatic stress disorder (PTSD) diagnoses or severity levels, and (2) whether the Capture and Rumination, Functional Avoidance, and Impaired Executive Control (CaR-FA-X) model applies to OGM in BPD. The study found that OGM in BPD was associated with age but not MDD, PTSD, BPD severity, trauma exposure, or the CaR-FA-X variables. OGM specificity was higher in B

8 Surprising Reasons To Meditate 40 Minutes A Day That Can Change Your Life.pptxHolistified Wellness
We’re talking about Vedic Meditation, a form of meditation that has been around for at least 5,000 years. Back then, the people who lived in the Indus Valley, now known as India and Pakistan, practised meditation as a fundamental part of daily life. This knowledge that has given us yoga and Ayurveda, was known as Veda, hence the name Vedic. And though there are some written records, the practice has been passed down verbally from generation to generation.

Overall life span (LS) was 1671.7±1721.6 days and cumulative 5YS reached 62.4%, 10 years – 50.4%, 20 years – 44.6%. 94 LCP lived more than 5 years without cancer (LS=2958.6±1723.6 days), 22 – more than 10 years (LS=5571±1841.8 days). 67 LCP died because of LC (LS=471.9±344 days). AT significantly improved 5YS (68% vs. 53.7%) (P=0.028 by log-rank test). Cox modeling displayed that 5YS of LCP significantly depended on: N0-N12, T3-4, blood cell circuit, cell ratio factors (ratio between cancer cells-CC and blood cells subpopulations), LC cell dynamics, recalcification time, heparin tolerance, prothrombin index, protein, AT, procedure type (P=0.000-0.031). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and N0-12 (rank=1), thrombocytes/CC (rank=2), segmented neutrophils/CC (3), eosinophils/CC (4), erythrocytes/CC (5), healthy cells/CC (6), lymphocytes/CC (7), stick neutrophils/CC (8), leucocytes/CC (9), monocytes/CC (10). Correct prediction of 5YS was 100% by neural networks computing (error=0.000; area under ROC curve=1.0).

(Hartman & Rothbart, 2001)
13. General Conclusions
Stable negative correlations between EC-scales and BPD complaints, even when
controlled for depression severity.
Both studies suggest that discrepancy is an important element, thereby supporting
the idea that high discrepant information leads to a reallocation of resources
towards the self (self-coherence), as is suggested by models on autobiographical
memory organisation.
Study 1: EC(ACT) mediates between BPD complaints and vantage perspective,
suggesting that BPD complaints will ameliorate from therapeutic strategies as MBCT,
ACT, MBT, and exposure.
Note: community sample! No data on trauma/medication/…
