T cells from strain 2 would respond to strain 13 cells due to alloreactivity between the different strains. B cells were used as target cells in Morrison and Braciale's experiments because B cells express both class I and class II MHC, allowing them to distinguish between the endogenous and exogenous antigen processing pathways using class I and class II MHC restricted T cells. Any cell type from the same animal could have been used but B cells are easy to obtain from blood.
