Fatty Acid Metabolism
Fatty Acids Roles
• A fatty acid contains a long hydrocarbon chain
and a terminal carboxylate group.
• Fatty acids are building blocks of
phospholipids and glycolipids.
• Many proteins are modified by the covalent
attachment of fatty acids.
• Fatty acids are fuel molecules.
• Fatty acid derivatives serve as hormones and
intracellular messengers.
Triacylglycerols Are Highly
Concentrated Energy Stores
• Since they are reduced and anhydrous.
• The yield from the complete oxidation of fatty
acids is about 9 kcal g-1 (38 kJ g-1), in contrast
with about 4 kcal g-1 (17 kJ g-1) for
carbohydrates and proteins.
• The major site of accumulation of
triacylglycerols is the cytoplasm of adipose
cells.
Dietary Lipids Are Digested by
Pancreatic Lipases
• Most lipids are ingested in the form of
triacylglycerols
• But must be degraded to fatty acids for
absorption across the intestinal epithelium.
• Triacylglycerols in the intestinal lumen are
incorporated into micelles formed with the aid
of bile salts
Dietary Lipids Are Transported in
Chylomicrons
• In the intestinal mucosal cells, the
triacylglycerols are resynthesized from fatty
acids and monoacylglycerols.

The Utilization of Fatty Acids as Fuel
Requires Three Stages of Processing
• Adipose tissues major reservoir.
• 3 steps for utilizations of fuel molecule.
– Mobilization (degradation of triacylglycerols and
transportation with the aid of albumin to required
tissues)
– Activation & transportation to mitochondria
– Oxidation of fatty acids to yield acetyl CoA
Triacylglycerols Are Hydrolyzed by
Cyclic AMP-Regulated Lipases
• Lipolysis…….. hydrolysis of triacylglycerols by
lipases
• Hormones inducing lipolysis….. epinephrine,
norepinephrine, glucagon, and
adrenocorticotropic hormones
• Insulin inhibitor of lipolysis

Fatty Acids Are Linked to Coenzyme A
Before They Are Oxidized
• In the outer mitochondrial membrane fatty
acids must be activated.
• Adenosine triphosphate (ATP) drives the
formation of a thioester linkage between the
carboxyl group of a fatty acid and the
sulfhydryl group of CoA.
Carnitine Carries Long-Chain Activated
Fatty Acids into the Matrix
Acetyl CoA, NADH, and FADH2 Are
Generated in Each Round of Fatty Acid
Oxidation
• 4 reactions during degradation of activated fatty
acids
– Oxidation by FAD
– Hydration
– Oxidation by NAD+
– Thiolysis by CoA
• The fatty acyl chain is shortened by two carbon
atoms as a result of these reactions, and FADH2,
NADH, and acetyl CoA are generated.
• The first reaction in each round of degradation
is the oxidation of acyl CoA by an acyl CoA
dehydrogenase to give an enoyl CoA with a
trans double bond between C-2 and C-3.
• The final step is the cleavage of 3-ketoacyl CoA
by the thiol group of a second molecule of
CoA, which yields acetyl CoA and an acyl CoA
shortened by two carbon atoms. This thiolytic
cleavage is catalyzed by b-ketothiolase.
The Complete Oxidation of Palmitate
Yields 106 Molecules of ATP
Fatty Acids Are Synthesized and
Degraded by Different Pathways
• Synthesis takes place in the cytosol, in
contrast with degradation, which takes place
primarily in the mitochondrial matrix.
• Intermediates in fatty acid synthesis are
covalently linked to the sulfhydryl groups of an
acyl carrier protein (ACP),
• Whereas intermediates in fatty acid
breakdown are covalently attached to the
sulfhydryl group of coenzyme A.
• The enzymes of fatty acid synthesis in higher
organisms are joined in a single polypeptide
chain called fatty acid synthase. In contrast,
the degradative enzymes do not seem to be
associated.
• The growing fatty acid chain is elongated by
the sequential addition of two-carbon units
derived from acetyl CoA.
• The reductant in fatty acid synthesis is NADPH,
whereas the oxidants in fatty acid degradation
are NAD + and FAD.
• Elongation by the fatty acid synthase complex
stops on formation of palmitate (C16). Further
elongation and the insertion of double bonds
are carried out by other enzyme systems.
