Nucleostemin



Nucleostemin is expressed in nucleoli of CNS stem cells,
 mbryonic stem cells and several cancer cells.

Nucleostemin expression decreases rapidly prior to
neuronal differentiation of CNS stem cells.

 he N-terminal basic domain specifies the p53 interaction
 nd growth regulatory functions.

考察
ヌ ク レ オ ス テ ミ ン の 発 現 量 を 減 少 さ せ る と 、 p14 ARF と
mdm2 タ ン パ ク 質 量 の 減 少 と 、 p53 タ ン パ ク 質 量 の 増 加 が
認められたことから、ヌクレオステミンは p53 制御経路上
で機能していることが示唆された。
細 胞 老 化 の M1 期 は 、 テ ロ メ ア の 短 縮 を 察 知 し た ATM に
よる p53 の安定化によって誘起される細胞周期の停止であ
る と考えられている。本実験から、テロメアの短縮に加え
て 、 ヌ ク レ オ ス テ ミ ン 量 の 減 少 も M1 期 に お け る p53 の 安
定化に関与していることが示唆された。
ヒ ト骨髄間質細胞には寿命があることが、分化させた細胞
を 移植し臓器の機能回復を計る再生医療に役立てる際の大
き な障壁となっている。ヌクレオステミン遺伝子の導入に
よ って細胞周期を回転させ、多くの細胞数を得ることが可
能 となれば、再生医療への実用化に寄与できると考えられ
