➢ The combination of drugs produced superior survival benefit and prolonged the survival of animals.
➢ The combination of chosen drugs synergistically reduced proliferation of glioma cells and induced apoptosis in vitro (Staining- Acridine
Orange/Ethidium Bromide, Hoechst 33342, JC-I).
➢ The combination therapy decreased cellular proliferation and in tumor sections, as assessed by H&E and IHC (Ki-67- Proliferation, VEGF-
Angiogenic marker).
➢ The combinatorial treatment promoted loss of mitochondrial membrane potential, with an increase in the percentage of cells in the G0-phase
leading to cell cycle arrest and apoptosis (Confocal Imaging and Flow cytometry).
➢ The drug metabolites have permeabilized the blood brain barrier and reached the target site (LC-MS analysis).
➢ The combinatorial treatment has inhibited PI3K, Wnt and JAK/STAT pathways leading to apoptosis (qRT- PCR, Immunoblotting & ELISA).

51. Concluding Remarks- Repurposing is the future…
• GBM is a deadly primary parenchymal CNS neoplasm disease with very dismal prognosis.
• By bypassing time-consuming chemical optimization and toxicology testing in drug
development steps, repurposing of existing approved drugs can help to better manage GBM.
• As compounds need to be brain penetrant to have maximum efficacy in GBM, it is ideal to
select compounds that are known to pass the blood brain barrier and are not substrates of
efflux transporters.
• Furthermore, uncovering pathways controlled by each drug is essential for identifying
therapeutic combinations for treating GBM.
