Covalent Organic Frameworks (COFs) Linkers

Covalent organic frameworks (COFs) are crystalline organic frameworks consisting of a network
structure made of covalent bonds. COFs are classified as porous crystalline materials similar to
metal-organic frameworks (MOFs)/porous coordination polymers (PCPs) and zeolites. They include
2D COFs, which are constructed by stacking layers of 2D covalently bonded sheets, and 3D COFs,
which are constructed by 3D connected frameworks. COFs are expected to be used as molecular
storage or separation materials, catalysts, electronic materials, energy storage materials, battery
materials, and drug delivery materials, due to their porosity, crystallinity, and structural diversity.
COFs are designed and synthesized by combining monomers, also called linkers, according to

[Product No. B6255] is an air-stable divalent palladium(II) complex that
efficiently catalyzes Suzuki-Miyaura coupling reaction. The reaction is applicable to substrates with
heteroatom containing groups (such as heteroaryl chlorides, thiol, amino, alkoxy amino and alkoxy
groups) which can be catalyst poisons. In addition, the complex exhibits higher turnover
numbers (TONs).

Cubane-Containing Building Blocks: The Bioisostere of Benzene Ring

In the field of drug design, cubane is considered to be the bioisostere of benzene due to its
diagonal distance of the skeleton being almost the same as the diameter of benzene. The strategy
to introduce the cubane skeleton instead of the benzene ring has gathered much interest in recent
drug discovery. This is expected to improve solubility and pharmacokinetics by breaking the
planarity of the entire molecule. TCI offers a series of cubane compounds with various functional
groups [Product No. D5542, M3466, M3505, C3810, C3808, B6415] useful for derivatization, and
also can provide custom synthesis of various derivatives related to these compounds.
The term regenerative medicine refers to a next-generational field of medical techniques in which
cells are taken from patients, cultured, expanded, and returned to the patient in some manner in
order to regenerate tissues or promote wound healing. The type of stem cell perhaps most
frequently employed in these applications are known as induced pluripotent stem cells (iPSCs).
In recent years, organoids have come to the forefront as a model for the application of human
iPSCs in regenerative medicine. Organoids are in-vitro 3D cultured cell aggregates derivable from
both iPSCs as well as somatic stem cells which are not only capable of self-organization and long
term self-renewal, but which also exhibit similar morphology and function to the tissues from which
they were derived. This is achieved through the use of physical and biochemical cues, such as
extracellular matrix components and niche factors, which are able to recapitulate cells' natural
environment within living tissue.

DOTA Derivatives as Chelating Agents for Radioisotopes

A Thioamide for the Potent Inhibition of TGF-β Signaling

A 83-01 [Product No. A3324] is a thioamide which acts as a
potent inhibitor of the TGF-β type I receptor superfamily
activin-like kinases Activin-Like Kinase 4 (ALK4), 5 (ALK5), and
7 (ALK7), making it specific for the TGF-β/Activin A/Nodal
branch of the TGF-β signaling pathway. Inhibition with A 83-01
prevents phosphorylation of Smad2/3, which in turn leads to
attenuation of TGF-β-mediated growth inhibition and
epithelial-to-mesenchymal transition. A 83-01 is a common
component of organoid culture media; where it is used in
concert with other niche factors to promote self-differentiation
and self-assembly of stem cells into functionalized structures
resembling native structures in vivo.
Product Highlights
DOTA (1,4,7,10-tetraazacyclodecane-1,4,7,10-tetraacetic acid) is a chelating agent which consists
of a 12-membered ring containing nitrogen and can coordinate with various metal species. DOTA
is widely used in PET (Positron Emission Tomography) and MRI (Magnetic Resonance Imaging)
scans due to the high stability of its coordination compounds. Several DOTA derivatives are
available such as azido-mono-amide-DOTA [Product No. A3478] with an azido group,
DOTA-ADIBO [Product No. D6006] with an ADIBO group, and DOTA-NHS hexafluorophosphate
trifluoroacetate [Product No. D6007] with an NHS group, allowing for quick and easy labeling of
various probes. Examples in the literature include: The DOTA labeling of trastuzumab with and the
subsequent coordination of the radioisotope 67
Ga allowed researchers to examine the dynamics
of trastuzumab in a rat model. Additionally, the DOTA labeling of polyamido-amine (PAMAM) with
and the subsequent coordination of the radioisotope 68
Ga have been an effective strategy to
study the localization of PAMAM in a mouse model since PAMAM is believed to be effective in
transporting drugs throughout the body.

Mediator for Organic Electrochemical Reactions

Recently, organic electrochemical reactions have attracted much attention due to their high
functional group tolerance and high energy efficiency, and have been applied to the total
synthesis of natural products. TCNHPI [Product No. H1765] is utilized as a mediator in organic
electrochemical reactions. So far, the scalable allylic C-H bond oxidation of terpenes such as
valencene and steroids using TCNHPI as a mediator have been reported.
Bis(phenylsulfonyl) sulfide [Product No. B3288] enables the construction of thiophene rings or
sulfide bonds in rigid units by the reacting with a variety of nucleophiles such as aryl lithium under
mild conditions. It is therefore a useful reagent and a synthetic equivalent of sulfur dichloride (SCl2
),
which is highly toxic and difficult to handle. In addition, benzenesulfinic acid, a by-product of the
reaction, is soluble in water and can be easily removed by liquid separation. It has been reported
that the thienoacenes synthesized by using bis(phenylsulfonyl) sulfide are used as OFET materials.

Dr. Rambabu Chegondi has been selected for the prestigious SERB-Science and Technology
Award for Research (SERB-STAR) Award, 2022. This award is instituted by the SERB,
Government of India to recognize and reward outstanding performance of Principal
Investigators (PIs) of individual-centric SERB Projects with an “Excellent” grade.
Dr. Rambabu Chegondi was chosen for the award by SERB-STAR Award Selection Committee
among the selected 18 Awardees in different disciplines all over the nation based upon
outcome of completed project, his overall research accomplishments and quality of publications
among eligible nominations.
Dr. Rambabu Chegondi gladly acknowledges TCI Chemicals as the preferred chemical supplier
for his research owing to the quality assurance and wide range of products available.

journals of high impact. Recently, he was awarded Dr. A V Rama Rao Young Scientist Award for the
year 2019 and Thieme Chemistry Journals Award 2023. Currently, he is also an Editorial Board
Member of “Organic Letters” a leading American Chemical Society Journal.

Dr. Maddi Sridhar Reddy has been selected for the prestigious SERB-Science and

month, a research grant of Rs. 10 lakhs per annum, and Rs.1 lakh per annum as overhead charges
for a period of three years.
Dr. Sridhar Reddy, received his M.Sc. degree from Osmania University and completed his
Ph.D. in Organic Chemistry from CSIR-IICT, Hyderabad. Later, he went to The University of
Sherbrooke, Canada for postdoctoral studies. After returning from Canada, he joined CSIR-CDRI,
Lucknow as a Senior Scientist and later on moved to CSIR-IICT in 2017. Dr. Sridhar Reddy
published more than 70 research publications in peer-reviewed international journals of high
impact. For his endeavors, he received the prestigious Thieme Young Scientist Award for the year
2015, CDRI incentive Award for the year 2016, AVRA Young Scientist Award for the year 2017 along
with SERB-STAR award for the year 2022.
He has used several metal-based catalysts purchased from TCI for executing
many of his projects related to this award. Some bioactive molecules used for late stage
functionalizations were also purchased from TCI.
He appreciates the prompt response by TCI, helped him get through his projects on time and in a
successful manner.

  • 5. 05 TCI’s Featured Products Covalent Organic Frameworks (COFs) Linkers ........................................................................................................................... Covalent organic frameworks (COFs) are crystalline organic frameworks consisting of a network structure made of covalent bonds. COFs are classified as porous crystalline materials similar to metal-organic frameworks (MOFs)/porous coordination polymers (PCPs) and zeolites. They include 2D COFs, which are constructed by stacking layers of 2D covalently bonded sheets, and 3D COFs, which are constructed by 3D connected frameworks. COFs are expected to be used as molecular storage or separation materials, catalysts, electronic materials, energy storage materials, battery materials, and drug delivery materials, due to their porosity, crystallinity, and structural diversity. COFs are designed and synthesized by combining monomers, also called linkers, according to intended topology. In the field of drug design, cubane is considered to be the bioisostere of benzene due to its diagonal distance of the skeleton being almost the same as the diameter of benzene. The strategy to introduce the cubane skeleton instead of the benzene ring has gathered much interest in recent drug discovery. This is expected to improve solubility and pharmacokinetics by breaking the planarity of the entire molecule.
