TOPIC:
PROBE DESIGNING WITH
SIGNAL MOLECULE
2. WHAT IS PROBE ?
A labelled DNA or RNA molecule.
They are single stranded.
Used to search sequence of interest from the mixture of
nucleic acid.
Important : Probe is a complementary to the sequence of
interest.
3. TYPES OF PROBES :
Enzymatic probe.
These probes detect target proteins using enzymes for detection.
HOW THIS PROBE WORK?
1. Enzyme are conjugated to another protein (eg : biotin)
2. The protein binds to target molecules.
3. A substrate of the specific enzyme is added.(DAB).
4. A visible reaction is formed.( It gives blue colour)
4. TYPES OF PROBES :
Enzymatic probe.
Florescence probe.
Radioactive probe.
Fluorescent molecules, also called fluorophores
or fluors responds distinctly to light and are
used to detect protein molecules /target
molecules.
A radioisotope, is used in the probe to label the
nucleic acid fragments of our interest.
