1-3-2. Victory of 2008, 18-years-old congenital blind man opened eyes
In April 2008, doctors at Moorefileds Eye Hospital in London, based on their gene
therapy skills and experience, tried a gene therapy for a man who lost his sight
completely and who could hardly see day or night, and finally he opened his eyes.
This is a huge event and a holy grail of science and technology. This is the
regenerating of photoreceptor cells of which their own functions were dead, and
the main character is a 18-years-old named Steven Howarth. As a result, he became
the first normal person in the world to walk on the streets alone at night as well as
in the daytime.
Howarth was the third blind man who had undergone gene therapy at the hospital.
After four months of surgery, doctors were all surprised at the fast recovery of
Howarth’s eye, greatly encouraged, and hoped that the future of gene therapy
would be bright for all the congenital blind1).

Dr. Robin Ali, who led the surgery, said, "I am all excited about the success of
gene transfer technology for three patients since 2007. I also cannot conceal
fear and excitement about the progress today. Also, I cannot sleep well if we
think about science and technology to be achieved in the future.“
Howarth, who could not walk in day and night, now is walking alone at night
or at the subway station. He is watching guitar strings and playing guitar.
James Bainbridge, who participated in the surgery, said, “It's hugely rewarding
and exciting to see that this new treatment can have this impact on a person's
quality of life.“ Howarth said, "I was really in pain, but not now, I feel like
throwing off a big burden, I think I was born into a new life.“
The study was funded by the Department of Health in the UK and the results
were published online in the New England Journal of Medicine on 27 April
2008, entitled "Effect of Gene Therapy on Visual Function in Leber's Congenital
Amaurosis”2) .

How did they treat it? - Ali team members, with controlling the promoter of
DNA, inserted the complementary DNA of RPE65 into adeno-associated
virus(AAV), put it in a needle plastic tube as cannula(kæ’njʊlə), and injected it into
the pigment layer. At this time, the complementary DNA of RPE65 as cDNA is
carried into the pigment layer through virus(AAV) as a vector, finally cDNA as a
newly injected gene is expressed in the pigment layer.
In the past, retroviruses have been used, but retroviruses remain to cause
cancer cells and other diseases. Recently, adenovirus, a kind of cold virus, has
been used because it comes out within one week after delivery of cDNA.
Complementarity means that RPE65 in the pigment layer is mutated and
replaced by a normal gene. A normal gene expression means that
photoreceptor cells can be regenerated and finally detect lights and colors,
thus the eye opens.

1-3-3. Future therapy & treatment - Now, gene therapy is popular as
described above with using a needle plastic tube as cannula and virus as a
vector, and the 18-years-old Howarth's vision is recovered within four months
after gene injection, but think that technology is getting more and more
innovative way year by year.
Within 10 years, we will inject dozens of genes, not one gene, and your eyes
will be opened in two months. In twenty years, your eyes will be opened in
one month. In 30 years, your eyes will be opened in one day. After 200 years,
we will enter a world where we inject genes through a light. In addition, we
will inject four bases that make up the gene, and also when we enter the
Aquarius(əkwέəriəs)(AD 2,100~AD 4,259), most of the blind may open their eyes
by injecting atoms.
The words of the Bible that Jesus taught us are to be that: "Someday, we shall
discover the knowledge of Jesus such as the science and technology that Jesus
healed all the blind with just saying ‘the word’ and touching eyes with spit
or saliva(səla’ɪvə)."
