Soltus Tablets (Generic Amisulpride Tablets)
Warnings
Neuroleptic Malignant Syndrome, a potentially fatal syndrome, characterized
by hyperthermia, muscle rigidity, autonomic instability, altered consciousness
and elevated CPK, may occur. In the event of hyperthermia, particularly with
high daily doses, all antipsychotic drugs including Soltus tablets must be
stopped.
Amisulpride induces a dose-dependent prolongation of the QT interval. This
effect, known to potentiate the risk of serious ventricular arrhythmias,
particularly of the torsades de pointes type, is enhanced in patients with
bradycardia, hypokalemia, or congenital or acquired prolonged QT interval
(combination with a medicinal product which prolongs the QTc interval)
In randomized, placebo-controlled clinical studies in elderly patients with
dementia and treated with certain atypical antipsychotics agents, a threefold
risk of stroke was observed versus placebo.

Soltus Tablets (Generic Amisulpride Tablets)
Warnings
Cases of venous thromboembolism (VTE) have been reported with antipsychotic
drugs. Since patients treated with antipsychotic drugs often present acquired risk
factors for VTE, any potential risk factors for VTE must be identified before and
during treatment with and preventive measures should be taken.
Amisulpride may increase prolactin levels. Therefore, caution should be exercised and
patients with a history or a family history of breast cancer should be closely monitored
during Amisulpride therapy.
Hyperglycemia has been reported in patients treated with some atypical antipsychotic
agents, including Amisulpride, therefore patients with an established diagnosis of
diabetes mellitus or with risk factors for diabetes who are started on Amisulpride,
should get appropriate glycaemic monitoring.
Amisulpride is eliminated by the renal route. In cases of renal insufficiency, the dose of
Soltus tablets must be decreased and intermittent treatment should be considered.

Soltus Tablets (Generic Amisulpride Tablets)
Warnings
Amisulpride can lower the seizure threshold. Therefore patients with a history
of seizures should be closely monitored during therapy with Soltus tablets.
In elderly patients, Soltus therapy, like other neuroleptics, must be used with
special caution because of a possible risk of hypotension or sedation.
Withdrawal symptoms have been described after abrupt cessation of high
therapeutic doses of antipsychotic drugs.
Leukopenia, neutropenia and agranulocytosis have been reported with
antipsychotics, including Soltus tablets. Unexplained infections or fever may
be evidence of blood dyscrasia and requires immediate haematological
investigation.
Caution should be also exercised when prescribing Soltus tablets to patients
with Parkinson’s disease since it may cause worsening of the disease.

Overdose
Exaggeration of the known pharmacological and adverse effects of
Amisulpride (Soltus Tablets) have been observed. These could
include drowsiness, sedation, hypotension, extrapyramidal
symptoms and coma. Fatal outcomes have been reported mainly in

Soltus Tablets (Generic Amisulpride Tablets)
Overdose
There is no specific antidote to Amisulpride. Appropriate
supportive measure should therefore be instituted: close
supervision of vital functions and, because of the risk of
prolongation of QT interval, continuous cardiac monitoring
until the patient recovers.
In case severe extrapyramidal symptoms occur, anticholinergic
agents should be administered. Since Amisulpride is weakly
dialysed, haemodialysis is not recommended as a method of
elimination.

Soltus Tablets During Pregnancy
Amisulpride (Soltus tablets) has been classified by the US FDA as
Pregnancy Category C.
Very limited clinical data on pregnancies exposed to this medicine are
available therefore the safety of Amisulpride during human pregnancy
has not been established.
Use of the drug is not recommended during pregnancy unless the
benefits justify the potential risks. Neonates exposed to antipsychotics
(including Amisulpride tablets) during the third trimester of pregnancy
are at risk of adverse reactions including extrapyramidal and/or
withdrawal symptoms that could vary in severity and duration following
delivery.
