This study aimed to identify nucleolar proteins that are differentially expressed in leukemia cell lines. The researchers isolated nucleoli from acute myeloid leukemia and acute lymphoblastic leukemia cell lines. Proteomic analysis identified 36 proteins that differed between AML and ALL lines and 29 proteins between pre-B ALL and pre-T ALL lines. Western blot and immunofluorescence analysis showed higher nucleolar expression of prohibitin and TDP-43 in some leukemia lines compared to others. The results demonstrate differences in nucleolar protein composition between leukemia subtypes that could be exploited for leukemia identification and classification.
