This document introduces an experiment to test hemagglutination and complement-mediated lysis. Red blood cells (RBCs) are incubated with serum containing antibody and complement, or with heated serum lacking complement. Hemagglutination occurs when RBCs bind to antibody alone. Hemolysis occurs when RBCs bind antibody and complement, leading to complement activation and cell lysis. The experiment uses sheep RBCs (antigen) with rabbit antibody and serum (from immunized or normal rabbits) to test these reactions.
This document provides an introduction to an immunology laboratory exercise on bacterial agglutination and immunoprecipitation. It defines immune responses as activities that maintain host health by protecting against pathogens. Antibodies secreted by immune cells counteract antigens, and depending on the antigen, this reaction causes either agglutination (clumping of particulate antigens) or precipitation (falling out of soluble antigen-antibody complexes). The lab will use bacterial agglutination, where antibodies react with surface antigens on bacteria, causing clumping. It will also use immunoprecipitation, where antibodies react with soluble antigens, forming complexes that precipitate.
This document discusses antibody-mediated rejection (AMR) in organ transplantation and methods for detecting donor-specific antibodies (DSAs).
It first describes how AMR occurs when antibodies react with antigens on vascular endothelium, causing complement activation and vascular damage. Diagnosis requires histologic lesions, detection of C4d factor, and identifying DSAs in serum.
It then discusses different antibody detection methods like panel-reactive antibodies (PRA), Luminex single antigen beads, and crossmatch tests. Flow cytometric crossmatching is more sensitive than CDC crossmatch in detecting low-titer or non-complement-fixing antibodies. Preformed DSAs, especially against HLA Class I and II, increase rejection
The document discusses flow cytometry detectors, electronics, and data analysis. It describes how light detection systems like photodiodes and photomultiplier tubes (PMTs) work to convert photons into electrical signals. It also discusses data acquisition, where measurements from detectors are stored as numerical values for each event. The goals of data analysis are to count and discriminate cells, apply gates and regions of interest, and report results through frequency distributions, statistics, and compensation calculations.
arose- more preferred than agar
Agar has strong negative charge; Agarose has almost none (no charge) - interactions between gel and reactants are minimized.
In ouchterlony double diffusion, both antigen and antibody
allowed to diffuse in to the gel. This assay is frequently used
for comparing different antigen preparations. the method is
called double since the antigen and antibody are allowed to
migrate towards each other in a gel and a line of precipitation
is formed where the two reactants are meet. This precipitation
is highly specific and is used by people working with
diagnosis and protein detection technique.
