ABSTRACT
 The objective of present study was to develope gastro retentive drug delivery
system for anti-hypertensive drug propranolol hydrochloride.
 The biggest problem in oral drug delivery is low and erotic drug bioavailability, so
to increase bioavailability of drug it should be retain in stomach was investigated.
 Hydroxy propyl methyl cellulose (HPMC), Hydroxy propyl cellulose (HPC),
Xanthum gum was evaluated for their gel forming abilities as polymer .
 Propranolol has extensive first pass metabolism so their bioavailability is
low.Therefore to increase bioavailability it was decided to formulate floating tablet of
propranolol so that it remains in upper part of GIT for longer time.
 Evaluate the physical properties, in-vitro release and in-vivo behavior of
propranolol drug. Floating tablet was formulated with HPMC & HPC Polymer to
enhance the retention of the drug into stomach to enhance release thus bioavailability
increase.
 For making the tablet which retain in upper GI Tract it should be float on the gastric
juices so to make it float it should be low in density.
• Oral drug delivery has most widely used routes of administration among all the routes
of administration among all the routes , It is most convenient and extensively used.
• GRDDS : It is the control release drug delivery system possessing the ability of being
retained in stomach and release drug over extended period of time.
• They can help in optimizing oral controlled drug delivery of drugs having absorption
window continuously release drugs for prolong period of time,
Thus ensure optimal bioavailability.
• Gastric emptying occur during fasting as well as fed state.
• Gastric emptying studies revealed that orally administered controlled release dosage
form have two complications
- Short gastric residence time
- Unpredictable gastric emptying time
• GRDDS as novel approach aim to address these limitations by facilitating prolonged
drug retention in stomach thereby enhancing drug absorption and bioavailability.
This retention is achieved through various mechanisms such as Buoyancy, swelling,
mucoadhesion or other innovative stratergies.

2. Drug unstable in upper part of GIT
3. Insoluble in intestinal fluid
4. Drug with variable bioavailability
5. Drug with site specific absorption in window

help to design pharmaceutical dosage form with prolong gastric residence time , in
particular floating drug delivery system are discussed. (2006)
