▪ Embryo culture is a type of Tissue Culture technique.

3. ▪ WHAT IS EMBRYOCULTURE ?
Deals with sterile isolation and In- vitro growth of
zygotic immature/mature embryo into a plantlet.
In Somatic Embryogenesis technique we develop the
embryo into the plant with the help of Explant.
The part we select out of an living organism from
which we make the whole plant is called as Explant.
5. MATURE EMBRYO CULTURE
▪ The development stage is gained by embryo.
▪ Isolation of mature embryo from ripe seeds.
▪ Used when embryo doesn’t survives in-vivo(because
of seed dormancy chemical inhibitors or mechanical
resistance).

7. IMMATURE EMBRYO
CULTURE/ EMBRYO RESCUE
METHOD
▪ It is still in the development stage.
▪ Isolation of immature embryo from
unripe/hybrid/sterile seed which fails to develop.
▪ Used to rescue embryoof wide crosses or distant
hybridization (inter-specific and inter-generic cross)
9. PROCEDURE OF EMBRYO CULTURE
Pluck or take developed fruit
Surface sterilize with 0.01% tween 20 for 15 minutes followed by
washing with distilled water
Sterilize with 0.01% mercuric chloride (HgCl2) for 10-15 minutes,
followed by washing with distilled water
Break seed aseptically and isolate the embryo
Inoculate embryo on callus proliferating media
After four weeks of inoculation callus appears.
After 4 weeks, green callus and embryoids starts appearing
And after 8 weeks of inoculation, subculture on shoot regenerating
medium is to be done.
After 12 weeks shoot appears.

EMBRYOCULTURE
▪ Production of plant from embryo.
▪ Study the relationship between embryo and endosperm.
▪ Prevents Embryo abortion.
▪ Overcome seed dormancy.
▪ Sterile seed due to early ripening e.g. Peach, cherry,
apricot and plum.
▪ Used in scientific study as well
▪ Clonal propagation
