Duovir Tablets (Generic Lamivudine and Zidovudine Tablets)

Duovir tablets (Generic Lamivudine and
Zidovudine tablets), a combination of 2
nucleoside analogue reverse transcriptase
inhibitors, are indicated in combination with
other antiretroviral agents for the treatment of
HIV-1 infection.

Active Ingredient
The active ingredients present in Duovir tablets
are Lamivudine and Zidovudine. Each film-
coated Duovir tablet contains 150 mg of
Lamivudine and 300 mg of Zidovudine.

Lamivudine and Zidovudine Tablets are
available under various Drug trade names
that include Combivir, Duovir, Cytocom,
Lamda-Z, Lamuzid, Virocomb, Zidovex-L.

Uses of Duovir Tablets
Duovir tablets (Lamivudine and Zidovudine combination) are used in the
treatment of human immunodeficiency virus (HIV) infection. HIV is the
virus that causes acquired immune deficiency syndrome (AIDS).
Lamivudine and Zidovudine combination will not cure or prevent HIV
infection or the symptoms of AIDS. This medicine helps keep HIV from
reproducing, and appears to slow down the destruction of the immune
system. This may help delay the development of serious health problems
usually related to AIDS or HIV infection. Duovir tablets will not keep you
from spreading HIV to other people. People who receive this medicine may
continue to have other problems usually related to AIDS or HIV infection.

Duovir Tablets Dosage
The dosage of Duovir tablets (Generic Lamivudine and
Zidovudine tablets) is given below:
The recommended dosage of Duovir (Lamivudine and
Zidovudine Tablets) is to be adminisered orally twice daily, with
or without meals.
The recommended oral dose of Duovir tablets for HIV-1 infected
adults and adolescents weighing greater than or equal to 30 kg is
1 tablet (containing 150 mg of Lamivudine and 300 mg of
Zidovudine) twice daily.

The dosage of Duovir tablets (Generic Lamivudine and
Zidovudine tablets) is given below:
The recommended oral dosage of Duovir tablets for pediatric
patients who weigh greater than or equal to 30 kg and for whom
a solid oral dosage form is appropriate is 1 tablet administered
twice daily.
Since Duovir is a fixed-dose combination tablet, it must not be
prescribed for pediatric patients weighing less than 30 kg or for
patients requiring dosage adjustment.

Contraindications
Duovir tablets are contraindicated in patients with a
hypersensitivity to Lamivudine, Zidovudine or any other
component of this medication.
Zidovudine is contraindicated in patients with abnormally low
neutrophil counts. Duovir tablets are therefore contraindicated
in these patients.

Storage Instructions
Duovir Tablets (Generic Lamivudine and
Zidovudine Tablets) have to be stored at
controlled room temperature i.e. from 20°C to
25°C (68°F to 77°F) with excursions
permitted between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Hepatic decompensation, some fatal, has occurred in HIV-1/HCV
coinfected patients receiving combination antiretroviral therapy and
interferon alfa with/without Ribavirin. Discontinue Lamivudine and
Zidovudine tablets as medically appropriate and consider dose reduction.
or discontinuation of interferon alfa, Ribavirin, or both.
Exacerbation of anemia has been reported in HIV-1/HCV co-infected
patients receiving Ribavirin and Zidovudine. Coadministration of Ribavirin
and Zidovudine is not advised.
Pancreatitis: Use with caution in patients with a history of pancreatitis or
other significant risk factors for pancreatitis. Discontinue treatment as
clinically appropriate.
Immune reconstitution syndrome and redistribution/accumulation of body
fat have been reported in patients treated with combination antiretroviral
therapy.

Side Effects
Most commonly reported side effects (incidence greater
than or equal to 15%) in clinical trials of combination
Lamivudine and Zidovudine were headache, nausea,
malaise and fatigue, nasal signs and symptoms, diarrhea,
and cough.

Overdose
There is no known specific treatment for overdose with Lamivudine and
Zidovudine tablets. If overdose occurs, the patient should be monitored and
standard supportive treatment applied as required.
Lamivudine: Because a negligible amount of Lamivudine was removed via
(4-hour) hemodialysis, continuous ambulatory peritoneal dialysis, and
automated peritoneal dialysis, it is not known if continuous hemodialysis
would provide clinical benefit in a Lamivudine overdose event.
Zidovudine: Acute overdoses of Zidovudine have been reported in pediatric
patients and adults. These involved exposures up to 50 grams. No specific
symptoms or signs have been identified following acute overdosage with
Zidovudine apart from those listed as adverse events such as fatigue,
headache, vomiting, and occasional reports of hematological disturbances.
Patients recovered without permanent sequelae.

Duovir Tablets During Pregnancy
Lamivudine and Zidovudine combination (Duovir Tablets) is classified
by the US FDA as Pregnancy Category C.
There are no adequate and well-controlled studies of Lamivudine and
Zidovudine Tablets (Duovir) in pregnant women. Clinical trial data
demonstrate that maternal zidovudine treatment during pregnancy
reduces vertical transmission of HIV-1 infection to the fetus.
Animal reproduction studies performed with Lamivudine and Zidovudine
tablets showed increased embryo toxicity and fetal malformations
(Zidovudine),and increased embryo lethality (Lamivudine).
Duovir tablets should be used during pregnancy only if the potential
benefit justifies the potential risk to the fetus.
