1. Anti-oxidant, anti-bacterial, and anti-inflammatory effects of Phyllanthus urinaria L.

Abstract : Phyllanthus urinaria L. has been used for the anti-inflammatory drug in Chinese traditional medicine. The
present study evaluated anti-oxidative, anti-bacterial and anti-inflammatory effects of Phyllanthus urinaria L. ethanol
extract (PUE). The antibacterial activity of PUE was tested against Edwardsiella tarda (E. tarda), Streptococcus
parauberis (S.parauberis) and Streptococcus iniae (S.iniae) using paper disc diffusion method and observed the
Minimum inhibitory concentration (MIC) values. In order to investigate the potential anti-inflammatory effects of PUE,
the production of nitric oxide (NO), TNF-α, IL-1β and IL-6 were measured in LPS stimulated RAW 264.7 cells. PUE
inhibited the production of NO,TNF-α,IL-1β and IL-6 at a concentration of 120µg/ml of PUE by 42 %, 94%, 23% and
47% respectively. Western blot analysis revealed that PUE inhibited the expressions of inducible nitric oxide
synthase (iNOS) and cyclooxygenase-2 (COX-2) protein levels in LPS stimulated RAW 246.7 cells. Overall results
suggest that the anti-bacterial and anti-inflammatory effects of PUE can be applied for the development of natural
antibiotics to prevent bacterial diseases in fish farm.
1.DPPH Assay :
The antioxidant activity of the PUE showed an anti-oxidative
effect by increasing DPPH radical scavenging ability in a
dose dependent manner.
2. Cell viability :
Cytotoxicity was not observed in the cytotoxicity of RAW
264.7 cell of PUE at the concentration of 1 ~ 300 μg/mL.
However, the survival rate was about 40% at the concentratio
n of 500 μg / mL, and the cell activity was significantly decre
ased (p <0.0001) at the above concentrations.
3.Antibacterial activity:
Antibacterial activity test showed that the PUE marbles
extracted with 80% EtOH forms a clear zone around paper
discs against E. tarda, S. parauberis, and S. iniae.
MIC values were estimated to be 800µg/ml for E.tarda,
800 µg/ml for S.parauberis and 240µg/ml for S.iniae
respectively.

5.Western blot :

Conclusion
In Conclusion, the current study evaluated anti-oxidati
ve, anti-bacterial and anti-inflammatory effects of
PUE. Western blot analysis revealed that PUE
inhibited the expressions of iNOS and COX-2 in LPS
stimulated RAW 246.7 cells. Therefore, PUE can be
applied for the development of natural antibiotics to
prevent bacterial diseases in fish farm.
