10. HPLC CHROMATOGRAPHY
Is a type of chromatography that stands for
High Performance Liquid Chromatography
It has two phases, viz; Mobile and Stationary Phases
A form of Liquid Chromatography used to separate compounds that are dissolved in solution
HPLC instruments consist of a reservoir of mobile phase, a pump, an injector, a column,
detector and a data recording and processing device.
Compounds are separated by injecting a sample mixture onto the column
Separation is based on Partition and or adsorption of compounds towards stationary phase
and mobile phase depends upon the compounds affinity toward either phase.
11. Types of HPLC
Normal Phase HPLC
In this type, the Stationary Phase is POLAR while the Mobile
Phase is NON-POLAR.
Reverse Phase HPLC
Here, the Stationary is NON-POLAR while the Mobile Phase is
POLAR.
