2. INTRODUCTION
• Used to identify Anthrax bacillus in microscope.
• Actually a Staining technique.
• Employed for the presumptive diagnosis of anthrax in animals.
• Stains the polypeptide capsular material of anthrax bacilli that is composed
of poly-d–Glutamic Acid.
3. PROCEDURE
• Take blood from tip of ear and make a impression slide.
• This is stained with Poly Chrome Methylene Blue (PCMB) for 30 seconds.
• Examine under microscope.
• If the sample is positive, an amorphous purplish material is noticed around
the blue bacilli.
• This purplish material is capsule around blue bacilli body.
