3. • Gastro-retentive drug delivery system (GRDDS) has gained immense
popularity in the field of oral drug delivery recently.
• It’s widely employed approach to retain the dosage form in the stomach for
extended period of time and release the drug slowly that can address many
challenge associated with conventional oral delivery including poor
bioavailability.
• Different innovative approaches like magnetic field assisted gastro-
retention plug type swelling system, muco-adhesion techniques, floating
system with or without effervescence are being applied to fabricate
GRDDS.

system to float and work efficiently.
2.Requires present of food to delay gastric empting.
3.Drugs which undergo first pass metabolism may not be desirable
candidates for floating DDS since the slow gastric empting .
4.It will leads to alter systemic bioavailability.
5.Drugs having solubility or stability problems in the highly acidic
gastric environment or which are irritant to gastric mucosa cant
be formulated as GRDDS.

8. • Floating system can be classified into two categories each class has subclasses:
1. Effervescent
a. Gas generating system
b. Volatile liquid containing system
c. Inflatable GIDS
d. Intra gastric osmotically controlled drug delivery system
2.Non-Effervescent
a. Colloidal gel barrier system
b. Alginate beads
c. Hollow microspheres
d. Microporous compartment system.

• Intra gastric single layer floating tablets or hydrodynamically balanced
system (HBS).

• These system can be made to float in the stomach
becaus of floatation chamber which maybe
a vacuum or field with air or a harmless
gas, while drug reservoir is encapsulated
inside microporous compartment.

11. c. Inflatable gastrointestinal delivery system
• In this system an inflatable chamber is
incorporated which contains liquid ether
that evaporate at body temperature to
cause the chamber to inflate in the stomach.
• The system is fabricated by loading the inflatable
chamber with drug reservoir which can be
a drug impregnated polymeric matrix then
encapsulated in a gelatine capsule.

12. d. Intra gastric osmotically controlled drug delivery system
• It’s comprised of an osmatic pressure-controlled drug delivery device and an inflatable
floating support in biodegradable capsule.
• I stomach the capsule quickly disintegrates to release the intrinsic osmotically
controlled drug delivery device.
