High capacity chromatography resin for the capture and purification of monoclonal antibodies
Tosoh Bioscience, a provider of chromatographic solutions for the isolation and analysis of biomolecules, introduced a new Protein A chromatography resin designed for the purification of monoclonal antibodies (mAbs). It is well-suited for high capacity capturing of immunoglobulin out of high titer feedstock. TOYOPEARL AF-rProtein A HC-650F achieves 30% to 50% greater antibody adsorption than similar products. It exhibits dynamic binding capacities of greater than 70 g/L at residence times of 5 minutes and greater than 50 g/L at residence times of 2 minutes with feedstock titers from 1 g/L to more than 10 g/L.
The recombinant ligand that is linked to the well proven methacrylic polymer backbone of TOYOPEARL media has been engineered to maintain capacity even after repeated exposure to alkaline solution. Its multipoint attachment to the TOYOPEARL matrix further enhances chemical and thermal stability. In practice this pays off for a low level of Protein A leaching and also for a high resistance to alkaline solutions applied in cleaning-in-place (CIP) procedures. Due to its rigid polymer matrix TOYOPEARL AF-rProtein A HC-650F also provides excellent pressure flow characteristics.
Protein A resins constitute a substantial cost in state-of-the-art mAb purification processes. Factors such as operating cycles, capacity, and mAb titer can have an impact on total costs associated with mAb purification. The high capacity of the new TOYOPEARL AF-rProtein A HC-650F resin and its high alkaline resistance increase product throughput, reduce operating costs and increase manufacturing productivity.
Over the past decade, there have been a growing number of mAb candidates entering the clinical pipeline. This results in a large increase on the demand for analytical characterization. This seminar discusses advances in analytical method development with analytical run times below 10 minutes for all routine methods with intelligent, integrated chromatography workflows. Orbitrap technology has been established as the most powerful MS technology for protein characterization. How this can be incorporated into a complete workflow for bio-pharma analysis is also discussed.
Layer-by-layer (LbL) films have been produced with
poly(o-ethoxyaniline) (POEA), chitosan and chitosanpoly(
methacrylic acid) (CS-PMAA) nanoparticles. Because the
adsorption of LbL films depends on ionic interactions and Hbonding, optimized conditions had to be established for the growth of multilayer films. Unusually thick films were obtained for POEA and CS-PMAA, thus demonstrating the importance of using chitosan in the form of nanoparticles.

Besides other applications this is of special interest for the purification of plasma proteins.
Our team of 8 undergrads spent a summer conceiving, designing, constructing, and testing a new system for sensing and reporting alternative fuel source concentrations. We were lucky enough to be invited to the world championship jamboree at MIT in Cambridge, MA, and this was our presentation for our 20 minute talk. Polyethylene terephthalate (PET) is a thermoplastic polymer resin of the polyester family and is used in synthetic fibers, beverage, food and other liquid containers, thermoforming applications and engineering resins. Depending on its processing and thermal history, PET may exist both as an amorphous and as a semi-crystalline polymer. Polymerization is through a polycondensation reaction of the monomer ethylene terephthalate with water as the byproduct.The majority of the world's PET production is for synthetic fibers with bottle production accounting for around 30% of global demand. The polyester industry makes up about 18% of world polymer production and is third after polyethylene (PE) and polypropylene (PP). Mechanical recycling of PET becomes more and more economically valuable due to the growing environmental awareness of the (Western) customer. The possibility of using recycled PET which will come into contact with food creates the need to develop efficient and sensitive analytical methods applicable to the identification and quantification of contaminants from recycled PET, which would also evidence that this recycled material complies with the terms of legislation. The recycling processes of PET for direct food contact have to evidence efficiency of contaminant removal and ensure consumer protection.

Key aspects during downstream purification of biopharmaceutical drugs are purity and process yield. Therefore, the downstream process needs to be designed in a way that the final product which will eventually end up in the patient entails low levels of product- and process related impurities (e.g. high molecular weight aggregates) as well as process related contaminants (e.g. host cell protein levels). In addition to this, the process must be capable of clearing and inactivating viruses to ensure product safety. In this webinar, we will explore the benefits of adding excipients during downstream processing on protein stability, chromatographic performance and viral inactivation.
