There is no basic formula to make an
organoid, but three things are important:
Pluripotent (PSC)/Stem cells (ESC) à
growth factors-supplemented media à
3D Culture.
The growth factors are crucial to induce
the thyroid fate in the cells, and that is
very challenging.
Scientists identified BMP4 and FGF2 as
important activators of thyroid
differentiation pathways.
They showed that PSC are activated by
BMP4/FGF2 into thyroid progenitor cells.
Those cells can form Organoids when in
3D culture and in the presence of TSH.
Thyroid organoids were transplanted in
hypothyroid mouse and it thyroid function
was recovered.
