This document discusses developing liver pyruvate kinase inhibitors through structural guided optimization to treat metabolic associated fatty liver disease. It first introduces the problem of NAFLD and lack of approved pharmacological treatments. It then details selecting the JNK protein kinase target and enumerating potential inhibitor compounds through docking simulations. Several analogues were synthesized and top performers were tested, showing inhibition of JNK3 isoform and effects on reducing triacylglyceride levels and improving cell viability. The conclusion discusses overcoming challenges of structural complexity, off-target effects, and ensuring safety and reducing toxicity.

Our speaker, Dr. Fergus J. Couch of the Mayo Clinic, presents on the design of this NCI-funded project, which comprises the sequencing of 60,000 samples to assess the risk of breast cancer through association with targeted genes. The design and size of the study requires an accurate, robust and high-throughput sequencing method. The investigators are using a digital DNA sequencing approach from QIAGEN that incorporates molecular barcodes to tag and remove PCR duplicates and increase NGS assay sensitivity. The approach also uses proprietary chemistry that enables uniform sequencing to efficiently utilize sequencing power and deliver optimized results.

However, due to the complexity of the biofluid samples accurate and precise measurements can become challenging due to poor chromatographic peak shape, insufficient chromatographic resolution from matrix components and incompatible sample compositions.
Recent advancements in the field of super critical fluid chromatography (SFC) have lead to the development of sub 2 micron chromatographic separations coupled mass spectrometry operating under positive ESI mode. In the work presented here the applicability of SFC for both chiral and achiral bioanalysis is shown. The orthogonal separation selectivity compared with reversed-phase separations and tolerance for high organic sample compositions will be discussed. This work further presents a critical evaluation of the influence of mobile phase and make up flow modifiers on the sensitivity and selectivity of probe pharmaceuticals analyzed under SFC/ESI positive mode MS conditions.

AACC 2019 -- Calprotectin is a calcium-heterodimer protein which is abundant in the cytoplasm of neutrophils. This is a biomarker with good sensitivity and specificity in case of inflammatory Bowel disease (IBD) which is a chronic inflammatory gut. In case of IBD, neutrophils from the inflammatory area release calprotectin, which leads to its increased levels in stool samples. Calprotectin is measured in extracted stools. There are several extraction devices that are commercially available as well a manual weigh-in method, "Gold Standard" method. The homogeneity of stool sample and the neutrophil levels in the sample affect the precision of the results from the same stool sample. Our study compares 2 commercial stool extraction devices with the manual weigh-in method as well as the variability within each extraction method.

STATEMENT OF NEED
Mantle cell lymphoma (MCL) is a rare, aggressive B-cell non-Hodgkin lymphoma (NHL) accounting for 5% to 7% of all lymphomas. Its prognosis ranges from indolent disease that does not require treatment for years to very aggressive disease, which is associated with poor survival (Silkenstedt et al, 2021). Typically, MCL is diagnosed at advanced stage and in older patients who cannot tolerate intensive therapy (NCCN, 2022). Although recent advances have slightly increased remission rates, recurrence and relapse remain very common, leading to a median overall survival between 3 and 6 years (LLS, 2021). Though there are several effective options, progress is still needed towards establishing an accepted frontline approach for MCL (Castellino et al, 2022). Treatment selection and management of MCL are complicated by the heterogeneity of prognosis, advanced age and comorbidities of patients, and lack of an established standard approach for treatment, making it vital that clinicians be familiar with the latest research and advances in this area. In this activity chaired by Michael Wang, MD, Professor in the Department of Lymphoma & Myeloma at MD Anderson Cancer Center, expert faculty will discuss prognostic factors informing treatment, the promising results of recent trials in new therapeutic approaches, and the implications of treatment resistance in therapeutic selection for MCL.
