       Conducted discovery research to identify and validate new targets for Obesity and Metabolic
       Disease group.

        Implemented and populated a repository for molecular biology reagents that include, plasmid
       clones, maxipreps and glycerol stocks of DNA clones and sequencing primers.
               Expedite the tracking for every reagent by creating excel based database which also
               contained relevant information for each of the reagent.

       Manufactured 300-400 Plasmid clones using basic molecular lab techniques.
              Constructed plasmid clones that contained target gene of interest by following regular
              cloning strategies.
              Amplified cloned plasmid DNA through bacterial transformation and performed high
              throughput DNA isolation using Vacuum manifold protocol and 96well format DNA
              isolation.
              Optimized the screening of positive clones through restriction digestion analysis and
              DNA sequencing.

       Analyzed protein expression after transfecting them in mammalian cell system, through
       immunochemical assays like SDS PAGE and western blotting.

       Characterized antibodies and used them in developing functional cell based assays to screen
       activity and localization for new targets.

           pET duet I and thereafter analyzed its protein expression in E.coli BL21 DE3 cells.

“Identification of unknown cDNA by inducing and analyzing protein expression in E.coli BL21 DE3 cells
as ACMPK gene that encodes for Ca2+/calmodulin (CaM) – dependent multi functional protein kinase”

       •   Characterized and identified the unknown cloned cDNA through restriction enzyme digestion as ACMPK
           gene.
       •   Amplified the cloned cDNA using E.coli DH5α cells and after inducing protein expression through E.coli
           BL21 cells isolated and purified the corresponding CaMK protein by Ni Affinity Column
           Chromatography.
