Vacuoles play several important roles in plant cells. They can contain multiple compartments to carry out different functions. Transporters in the vacuolar membrane, including proton pumps, aquaporins, and various channels, regulate the transport of substances like sugars, acids, and secondary metabolites into and out of the vacuole. This transport affects processes like fruit growth, taste, and quality. Proton pumps generate a proton gradient across the membrane that drives secondary active transport. Aquaporins transport water, while channels transport sugars, organic acids, and other compounds to store or sequester them in the vacuole.

large vacuoles, with different functions.
 Remarkably, even a single vacuole may be
subdivided and full fil several functions

Proton pumps V-ATPase and V-PPase generate an electrochemical
gradient (pH gradient and ΔΨ) across the vacuolar membrane.
 1) V-Ppase :
It consists of a single kind of polypeptide.
V-PPase appears to be the major proton pump.
It has a central role during the early stages of
cell expansion & cell division stage.
 2) V-ATPase :
It is a multisubunit enzyme complex comprising more than ten
different subunits.
V-ATPase allows a steeper pH gradient between the cytosol and the
vacuole.

Aquaporin's
 Water is transported by aquaporins in the
vacuolar membrane.
 Aquaporin is also called water channel or
major intrinsic protein (MIP).
 The aquaporin family is classified into four
subfamilies based on their sequence similarity:
1 : Plasma membrane intrinsic protein (PIP)
2 : Tonoplast intrinsic protein (TIP)
3 : NOD26-like intrinsic protein (NIP)
4 : Small basic intrinsic protein (SIP).

 Another factor determining acidity is the amount and type of
organic acids. The main organic acids in fruits are malate (e.g.
apple and pear), citrate (citrus) and tartrate (e.g. grape berry).
 Thus organic acids have a role as buffers and stabilize the pH.
 Also, each organic acid has a specific taste that contributes to the
overall taste of fruits.
 A first step was made by identifying the malate transporter
AttDT in Arabidopsis.
