Abstract Natural active molecules are key sources of modern innovative drugs. Particularly, a great amount of natural active molecules have been reported to possess promising therapeutic effects on inflammatory diseases, including asthma, rheumatoid arthritis, hepatitis, enteritis, metabolic disorders and neurodegenerative diseases. However, these natural active molecules with various molecular structures usually exert anti-inflammatory effects through diversiform pharmacological mechanisms, which is necessary to be summarized systematically. In this review, we introduced the current major anti-inflammatory natural active molecules based on their chemical structures, and discussed their pharmacological mechanisms including anti-inflammatory molecular signaling pathways and potential target proteins, which providing a referential significance on the development of novel anti-inflammatory drugs, and also revealing new therapeutic strategies for inflammatory diseases.

vivo anticonvulsant and antioxidant activities of Epilobium hirsutum, which
has been traditionally used in the treatment of epilepsy by local people of
Turkey. In vitro studies revealed that the extract contained a pronounced
amount of phenolics (206.3±0.9 mg Gallic acid Eq/g extract) and exhibited
significant levels of antioxidant (FRAP; 6226 µmol Fe2+/g extract, ORAC;
6593 µmol Trolox Eq/g extract, DPPH; IC50:33.8 ug/mL and metal chelation;
IC50:114 ug/mL) and anticonvulsant (AChE; IC50:71.2 ug/mL, BChE; IC50:92.5
ug/mL, GABA-T; IC50:94.7 ug/mL) activities. In vivo studies shown that the
the extract exhibited high anticonvulsant activities. In addition, the extracts
regulated the behavior, locomotion, and mental activities of the mice tested.
Biochemical evaluation of the brain tissue revealed that the extract inhibited
the production of MDA and stimulated the increase of antioxidant enzyme
levels, which suggest the possible antioxidative role of the extract that worked
as neuroprotective agents by scarfing the free radicals produced through PTZ
seizure inducer and attenuate convulsions. Moreover the extract regulated
serum biochemical parameters, total antioxidants, total oxidant, and ischemia modified albumin levels. Chromatographic studies were revealed that gallic
acid principally might be the major contributor of anticonvulsant and
antioxidant activities with the additive contributions of fatty acids and mineral
compounds. Findings obtained from this study partially justified the traditional
use of Epilobium hirsutum in the treatment of epilepsy and suggest potential
use of the extract as an industrial or pharmaceutical agent.

for its treatment only provide palliative effect, as
cure for the disease still remains elusive. Jobelyn®
(JB), a potent anti-oxidant and anti-inflammatory
dietary supplement obtained from Sorghum bicolor,
has been claimed to relieve arthritic pain. Thus, this
study was designed to evaluate its effect on
inflammatory and biochemical changes as well as
neurobehavioural deficits associated with complete
Freund-adjuvant (CFA)-induced arthritis in mice.

8. The effect of JB (50, 100 and200 mg/kg) on
inflammatory oedema, neurobehavioural
deficits, levels of biomarkers of oxidative
stress and inflammatory cytokines (tumor
necrosis factor-alpha and interleukin-6)
induced by 0.1 mL of CFA (10 mg/mL)was
evaluated in male Swiss mice. Oral
administration of JB (100 and 200 mg/kg)
reduced inflammatory paw volume and
reversed sensori motor deficits induced by
CFA. JB also reduced pain episodes, anxiety
and depressive-like symptoms in CFA-mice.

11. The increased level of oxidative stress in the
joint and brain tissues of CFA mice was
reduced by JB. It also decreased tumor
necrosis factor-alpha and interleukin-6
levels induced by CFA inthe joint tissue of
mice. These findings suggest that Jobelyn®
attenuates inflammatory responses induced
by CFA in mice via inhibition of oxidative
stress and release of inflammatory
cytokines. The ability of JB to attenuate
CFA-induced nociception, sensori motor
deficits and depressive-like symptom
suggests it might improve the quality of life
of patients with arthritic conditions

14. Results: JB (50–200 mg/kg) given orally produced
a significant inhibition of acute inflammation
induced by carrageenan in rats. It reduced the
volume and number of leukocytes in inflammatory
fluid in the granuloma air pouch model of chronic
inflammation. It further decreased the levels of
MDA in the fluid suggesting antioxidant property.
JB elevated the concentrations of GSH in
inflammatory exudates indicating free radical
scavenging activity. It also significantly inhibited
RBC lysis caused by hypotonic medium, suggesting
membrane-stabilizing property.
