INTRODUCTION
 Ion exchange resins are insoluble polymers that
contain acidic or basic functional groups and have
the ability to exchange counter-ions within aqueous
solutions surrounding them.
 Available normally in the form of small (1-2 mm
diameter) beads, usually white or yellowish,
fabricated from an organic polymer substrate
backbone
 Ion exchange is an adsorption phenomenon where
the mechanism of adsorption is electrostatic.

FOR EXAMPLE:

o Electrostatic forces hold ions to charged functional
groups on the surface of the ion exchange resin.
o The adsorbed ions replace ions that are on the resin surface
on a 1:1 charge basis.
 Ion exchange fibers have been widely used in many
applications due to their
 high separation capacity
 fast ion exchange rate
 good electrical conductivity
 They have applications not only as drug carriers, but also in a
number of areas within formulations and drug delivery and
biomedical analysis.
 They are used for separation, purification, and
decontamination processes. E.g. Water Softening and Water
Purification.
 These resins are being used extensively in overcoming various
formulation-related problems including poor stability and poor
dissolution, for taste masking and as a powder processing aid 5
 Typical ion exchange resins are based on cross-linked
polystyrene
 The required active groups can be introduced after
polymerization, or substituted monomers can be used.
 For example, the cross-linking is often achieved by adding
0.5-25% of divinyl benzene to styrene at the polymerization
process
 Non-cross-linked polymers are used only rarely because
they are less stable

CHEMISTRY
 An ion exchange resin is a polymer (normally styrene) with
electrically charged sites at which one ion may replace
another.
 Natural soils, natural water, waste water, certain minerals
called zeolites , living materials like cell walls, cell
membranes and other structures have charges and can
undergo ion exchange phenomenon.

 Synthetic ion exchange resins are usually cast as porous beads
with considerable external and pore surface where ions can
attach
 The resins are prepared as spherical beads 0.5 to 1.0 mm in
diameter
 These appear solid even under the microscope, but on a
molecular scale the structure is quite open

 While there are numerous functional groups that have charge,
only a few are commonly used for man-made ion exchange
resins.
 These are:
 -COOH, which is weakly ionized to -COO¯
 -SO3H, which is strongly ionized to -SO3¯
 -NH2, which weakly attracts protons to form NH3+
 -secondary and tertiary amines that also attract protons weakly
 -NR3+, which has a strong, permanent charge (R stands for
some organic group)
 These groups are sufficient to allow selection of a resin with
either weak or strong positive or negative charge.

CLASSIFICATION

1.CATION EXCHANGE RESINS
 Exchangeable ions are positively charged
 Prepared by the copolymerization of styrene and divinyl
benzene and have sulfonic acid groups
(-SO3H) introduced into most of the benzene rings.
 Mechanism

A. STRONG ACID CATION
EXCHANGE RESIN
 Highly ionized in both the acid (R-SO3H) and salt (RSO3Na)
form of the sulfonic acid group (-SO3H)
2(R-SO3H ) + NiCl2 → (R-SO4 )Ni + 2HCl
 Exchange capacity of strong acid resins is independent of the
solution pH.
 Used in the hydrogen form for complete deionization
 Used in the sodium form for water softening (calcium and
magnesium removal).
 After exhaustion, the resin is converted back to the hydrogen
form (regenerated) by contact with a strong acid solution, or the
resin can be convened to the Na form with a NaCl solution

 Ionizable group is a carboxylic acid (COOH)
 The degree of dissociation is strongly influenced by the
solution pH.
 Limited capacity below a pH of 6.0

 Exchangeable ions are negatively charged
 Mechanism
Resin+ - ex - + A- → Resin+ - A- + ex-

A. STRONG BASE ANION EXCHANGE
RESINS
 Highly ionized and can be used over the entire pH range.
 Used in the hydroxide (OH) form for water deionzation
 React with anions in solution and can convert an acid solution
to pure water
R-NH3OH + HCl → R-NH3Cl + HOH
 Regeneration with concentrated sodium hydroxide (NaOH)
converts the exhausted resin to the OH form.

B. WEAK BASE ANION EXCHANGE
RESINS
 Degree of ionization is strongly influenced by pH.
 Exhibit minimum exchange capacity above a pH of 7.0.
 Does not have a OH ion form as does the strong base resin.
R-NH2 + HCl → R-NH3Cl
 Regeneration needs only to neutralize the absorbed acid

EXCHANGE RESINS
1. Cross-linking
 Amount of cross-linking depends on the proportions of
different monomers used in the polymerization step.
Properties that are interrelated with cross-linking
are:
a) Capacity
 The total capacity of an ion exchange resin is
defined as the total number of chemical
equivalents available for exchange per some unit
weight or unit volume of resin.
 The more highly cross-linked a resin, the more
difficult it becomes to introduce additional
functional groups.

 Sulfonation is carried out after the cross-linking has been
completed and the Sulfonic acid groups (-SO3H) are
introduced inside the resin particle as well as over its surface.
b)Moisture Content
 Sulfonic acid groups (-SO3H) attract water, and this water is
tenaciously held inside each resin particle.
 The quaternary ammonium groups of the anion resins also
behave in a similar manner.
c) Equilibration Rate
 Cross-linkage has a definite influence on the time required
for an ion to reach equilibrium.
 The larger the ion or molecule diffusing into an ion exchange
particle, or the more highly cross-linked the polymer, the
longer the time required to reach equilibrium conditions.

2. AVAILABLE CAPACITY
 The capacity of an ion exchange is a quantitative measure of
its ability to take up exchangeable counter ions and it is
therefore of major importance.
a)Acid-Base Strength
 The acid or base strength of an exchanger is dependent on the
various ionogenic groups incorporated into the resin.
 Resin-containing sulfonic , phosphoric and carboxylic acid
exchanger groups have approximate pKa values of <1, 2-3
and 4-6, respectively.
 Anionic exchangers are quaternary, tertiary, or secondary
ammonium groups having apparent pKa values of >13, 7-9 or
5-9, respectively 19
b) Selectivity of the Resins for the Counter-Ion
 Selectivity depend mainly on the relative change and the
ionic radius of the (hydrated) ion competing for an
exchange site.
The extent of adsorption increases with -
 The counter ion that in addition to forming a normal ionic
bond with the functional group of an exchanger, also
interacts through the influence of van der waal forces with
the resin matrix.
 The counter ion at least affected by complex formation
with its co-ion or non-exchange ion.
 The counter ions that induce the greater polarization. These
factors, together with the effect of the size and charge of an
ion on exhibiting certain selectivity toward a resin, are at
best only general rules, and as a consequence there are
many exceptions to them.

TASTE MASKING
 Excessive bitterness of APIs in oral formulations is the major
taste problem
 Bitterness of formulations influence selection by physicians
and affect patient compliance
 Masking of the unpleasant taste of a drug improves
compliance and product value
 Numerous available taste-masking methods, ion exchange
resins are inexpensive
 Simple, rapid and cost-effective method of taste masking

Ion exchange resins used for masking the unpleasant taste of drugs
RAPID DISSOLUTION
 Ion exchange drug resinate complexes have a faster rate of
dissolution.
 Ion exchange resin matrices are hydrophilic
 Allow aqueous solutions to enter the resin structure, enhancing
the dissolution rate
 Each individual drug molecule is bound to a functional site of
the resin molecule
 Reduction of crystal lattice energy, thus enhancing the rate of
drug dissolution bound to resin

POWDER PROCESSING AID
 Hygroscopic drugs are susceptible to agglomeration due
to the presence of moisture
 Adsorption of such drugs onto ion exchange resins may
lead to a decrease in their hygroscopicity
 Resins have a uniform, macroreticular morphology
 Provide excellent flowability to the formulation

STABILITY
 Drug resinate is frequently more stable than the original drug.
 Vitamin b12 has a shelf-life of only a few months, but the
resinate is stable for more than two years.
 Nicotine it discolors quickly on exposure to air
 And light, but the resinate, used in nicotine chewing gums and
lozenges, is much more stable
 Ion exchange fibers provide a promising alternative to control
drug delivery and to store drugs that are degraded easily.

DELIQUESCENCE
 Deliquescence is defined as the conversion of a solid substance
into a liquid due to absorption of water vapor from the air
 Ion exchange resins may prove instrumental in solving the
problem of deliquescence of a drug by the formation of resinate
complexes
 Sodium valproate, a highly deliquescent drug
 After complexation with ion exchange resins showed free
flowing properties.
 The amount of water absorbed decreases with increasing
amount of valproate in the resinate.

DISINTEGRATION
 Ion exchange resins, has excellent swelling property when
immersed in water
 can be used as a tablet disintegrating agent

DRUG DELIVERY APPLICATIONS
1. Oral Drug Delivery
Modified Release: E.g.
• Sustained release suspension of diltiazem using strong
cation exchange resins for the preparation of microcapsules
by emulsion solvent evaporation method.
• The sustained release suspensions possessed good stability
with low drug leaching, and their release profiles are
unchanged when compared with the dried microcapsules for
120 days at 30 and 45°C.

2. Nasal Drug Delivery
Ion exchange resins to develop novel nasal formulations of
nicotine.
• A novel nasal formulation, in the form of a nicotine-
Amberlite resin complex powder, has been developed that
provided an optimal combined pulsatile and sustained plasma
nicotine profile for smoking cessation.
• Amberlite IRP69 and Amberlite IR120 are similar cationic
exchange materials with the same ion exchange capacity but
due to a smaller particle size range (10-150 micron),
Amberlite IRP69 had a better flow property and a better
adsorptive capacity than Amberlite IR120.
• The nicotine plasma profiles demonstrated that an initial
rapid peak plasma level of nicotine followed by a sustained
elevated level could be achieved by adjusting the ratio of free
to bound nicotine in the Amberlite powder formulation

3. Transdermal Drug Delivery:
• The model for transdermal delivery using ion exchange fibers
as controlling device was designed, and the main objective of
their study was to assess the rationality of the model.
• The release rates of ketoprofen from the carbopol-based gel
vehicles containing ion exchange fibers to which the ketoprofen
had been bound were determined across 0.22 μm micro porous
membrane.
• The fluctuation of the release rate of ketoprofen from the
vehicles was much lower compared with that of simple gels,
though the cumulative amount of ketoprofen delivery was less.
• Additional ions could increase the rate and extent of ketoprofen
delivery.

4. Ophthalmic Drug Delivery:
• The complexation of betaxolol hydrochloride, an
antiglaucoma agent with ion exchange resins.
• Drug resin particles are incorporated into the structured
vehicle, containing Carbomer 934P as a polymer, to
enhance the physical stability and ease of resuspendability
of the product.
• The 0.25% ophthalmic suspension of the drug showed an
increased bioavailability, and pharmaceutically, is an
elegant suspension product, which settles slowly, providing
uniform dosage and increased ocular comfort.

INTRODUCTION  Ion exchange resins are insoluble polymers that contain acidic or basic functional groups and have the ability to exchange counter-ions within aqueous solutions surrounding them.  Available normally in the form of small (1-2 mm diameter) beads, usually white or yellowish, fabricated from an organic polymer substrate backbone  Ion exchange is an adsorption phenomenon where the mechanism of adsorption is electrostatic. 3
  • 4. FOR EXAMPLE: 4 o Electrostatic forces hold ions to charged functional groups on the surface of the ion exchange resin. o The adsorbed ions replace ions that are on the resin surface on a 1:1 charge basis.
  • 5.  Ion exchange fibers have been widely used in many applications due to their  high separation capacity  fast ion exchange rate  good electrical conductivity  They have applications not only as drug carriers, but also in a number of areas within formulations and drug delivery and biomedical analysis.  They are used for separation, purification, and decontamination processes. E.g. Water Softening and Water Purification.  These resins are being used extensively in overcoming various formulation-related problems including poor stability and poor dissolution, for taste masking and as a powder processing aid 5
  • 6.  Typical ion exchange resins are based on cross-linked polystyrene  The required active groups can be introduced after polymerization, or substituted monomers can be used.  For example, the cross-linking is often achieved by adding 0.5-25% of divinyl benzene to styrene at the polymerization process  Non-cross-linked polymers are used only rarely because they are less stable 6

CHEMISTRY  An ion exchange resin is a polymer (normally styrene) with electrically charged sites at which one ion may replace another.  Natural soils, natural water, waste water, certain minerals called zeolites , living materials like cell walls, cell membranes and other structures have charges and can undergo ion exchange phenomenon. 7
  • 8.  Synthetic ion exchange resins are usually cast as porous beads with considerable external and pore surface where ions can attach  The resins are prepared as spherical beads 0.5 to 1.0 mm in diameter  These appear solid even under the microscope, but on a molecular scale the structure is quite open 8
  • 9.  While there are numerous functional groups that have charge, only a few are commonly used for man-made ion exchange resins.  These are:  -COOH, which is weakly ionized to -COO¯  -SO3H, which is strongly ionized to -SO3¯  -NH2, which weakly attracts protons to form NH3+  -secondary and tertiary amines that also attract protons weakly  -NR3+, which has a strong, permanent charge (R stands for some organic group)  These groups are sufficient to allow selection of a resin with either weak or strong positive or negative charge. 9
  • 11. 1.CATION EXCHANGE RESINS  Exchangeable ions are positively charged  Prepared by the copolymerization of styrene and divinyl benzene and have sulfonic acid groups (-SO3H) introduced into most of the benzene rings.  Mechanism Resin- - ex+ + C+ → Resin- - C+ +ex+ 11

STRONG ACID CATION EXCHANGE RESIN  Highly ionized in both the acid (R-SO3H) and salt (RSO3Na) form of the sulfonic acid group (-SO3H) 2(R-SO3H ) + NiCl2 → (R-SO4 )Ni + 2HCl  Exchange capacity of strong acid resins is independent of the solution pH.  Used in the hydrogen form for complete deionization  Used in the sodium form for water softening (calcium and magnesium removal).  After exhaustion, the resin is converted back to the hydrogen form (regenerated) by contact with a strong acid solution, or the resin can be convened to the Na form with a NaCl solution 12
  • 13. B. WEAK ACID CATION EXCHANGE RESINS  Ionizable group is a carboxylic acid (COOH)  The degree of dissociation is strongly influenced by the solution pH.  Limited capacity below a pH of 6.0 13
  • 14. 2. ANION EXCHANGE RESINS  Exchangeable ions are negatively charged  Mechanism Resin+ - ex - + A- → Resin+ - A- + ex- 14
  • 15. A. STRONG BASE ANION EXCHANGE RESINS  Highly ionized and can be used over the entire pH range.  Used in the hydroxide (OH) form for water deionzation  React with anions in solution and can convert an acid solution to pure water R-NH3OH + HCl → R-NH3Cl + HOH  Regeneration with concentrated sodium hydroxide (NaOH) converts the exhausted resin to the OH form. 15
  • 16. B. WEAK BASE ANION EXCHANGE RESINS  Degree of ionization is strongly influenced by pH.  Exhibit minimum exchange capacity above a pH of 7.0.  Does not have a OH ion form as does the strong base resin. R-NH2 + HCl → R-NH3Cl  Regeneration needs only to neutralize the absorbed acid 16
  • 17. PROPERTIES OF ION EXCHANGE RESINS 1. Cross-linking  Amount of cross-linking depends on the proportions of different monomers used in the polymerization step. Properties that are interrelated with cross-linking are: a) Capacity  The total capacity of an ion exchange resin is defined as the total number of chemical equivalents available for exchange per some unit weight or unit volume of resin.  The more highly cross-linked a resin, the more difficult it becomes to introduce additional functional groups. 17
  • 18.  Sulfonation is carried out after the cross-linking has been completed and the Sulfonic acid groups (-SO3H) are introduced inside the resin particle as well as over its surface. b)Moisture Content  Sulfonic acid groups (-SO3H) attract water, and this water is tenaciously held inside each resin particle.  The quaternary ammonium groups of the anion resins also behave in a similar manner. c) Equilibration Rate  Cross-linkage has a definite influence on the time required for an ion to reach equilibrium.  The larger the ion or molecule diffusing into an ion exchange particle, or the more highly cross-linked the polymer, the longer the time required to reach equilibrium conditions. 18

AVAILABLE CAPACITY  The capacity of an ion exchange is a quantitative measure of its ability to take up exchangeable counter ions and it is therefore of major importance. a)Acid-Base Strength  The acid or base strength of an exchanger is dependent on the various ionogenic groups incorporated into the resin.  Resin-containing sulfonic , phosphoric and carboxylic acid exchanger groups have approximate pKa values of <1, 2-3 and 4-6, respectively.  Anionic exchangers are quaternary, tertiary, or secondary ammonium groups having apparent pKa values of >13, 7-9 or 5-9, respectively 19
  • 20. b) Selectivity of the Resins for the Counter-Ion  Selectivity depend mainly on the relative change and the ionic radius of the (hydrated) ion competing for an exchange site. The extent of adsorption increases with -  The counter ion that in addition to forming a normal ionic bond with the functional group of an exchanger, also interacts through the influence of van der waal forces with the resin matrix.  The counter ion at least affected by complex formation with its co-ion or non-exchange ion.  The counter ions that induce the greater polarization. These factors, together with the effect of the size and charge of an ion on exhibiting certain selectivity toward a resin, are at best only general rules, and as a consequence there are many exceptions to them. 20

TASTE MASKING  Excessive bitterness of APIs in oral formulations is the major taste problem  Bitterness of formulations influence selection by physicians and affect patient compliance  Masking of the unpleasant taste of a drug improves compliance and product value  Numerous available taste-masking methods, ion exchange resins are inexpensive  Simple, rapid and cost-effective method of taste masking 22

Oral Drug Delivery Modified Release: E.g. • Sustained release suspension of diltiazem using strong cation exchange resins for the preparation of microcapsules by emulsion solvent evaporation method. • The sustained release suspensions possessed good stability with low drug leaching, and their release profiles are unchanged when compared with the dried microcapsules for 120 days at 30 and 45°C. 29

Nasal Drug Delivery Ion exchange resins to develop novel nasal formulations of nicotine. • A novel nasal formulation, in the form of a nicotine- Amberlite resin complex powder, has been developed that provided an optimal combined pulsatile and sustained plasma nicotine profile for smoking cessation. • Amberlite IRP69 and Amberlite IR120 are similar cationic exchange materials with the same ion exchange capacity but due to a smaller particle size range (10-150 micron), Amberlite IRP69 had a better flow property and a better adsorptive capacity than Amberlite IR120. • The nicotine plasma profiles demonstrated that an initial rapid peak plasma level of nicotine followed by a sustained elevated level could be achieved by adjusting the ratio of free to bound nicotine in the Amberlite powder formulation 30

Transdermal Drug Delivery: • The model for transdermal delivery using ion exchange fibers as controlling device was designed, and the main objective of their study was to assess the rationality of the model. • The release rates of ketoprofen from the carbopol-based gel vehicles containing ion exchange fibers to which the ketoprofen had been bound were determined across 0.22 μm micro porous membrane. • The fluctuation of the release rate of ketoprofen from the vehicles was much lower compared with that of simple gels, though the cumulative amount of ketoprofen delivery was less. • Additional ions could increase the rate and extent of ketoprofen delivery. 31

Ophthalmic Drug Delivery: • The complexation of betaxolol hydrochloride, an antiglaucoma agent with ion exchange resins. • Drug resin particles are incorporated into the structured vehicle, containing Carbomer 934P as a polymer, to enhance the physical stability and ease of resuspendability of the product. • The 0.25% ophthalmic suspension of the drug showed an increased bioavailability, and pharmaceutically, is an elegant suspension product, which settles slowly, providing uniform dosage and increased ocular comfort. 32
