This study used an electrotaxis assay to separate Caenorhabditis elegans nematodes into groups based on their crawling speed in response to an electric field. Nematodes that crawled more slowly had shorter lifespans, higher levels of protein damage, and lower heat shock resistance than faster nematodes. Gene expression analysis found that slow nematodes had higher transcript levels of heat shock genes, which correlated with their poorer stress response and shorter lifespan. The results suggest that accumulation of early-life damage leads to faster age-related deterioration and a shorter lifespan.

Cucumber mosaic virus was found for the first time infecting pot marigold (Calendula officinalis) plants in Serbia, causing chlorotic mottling and leaf deformation symptoms. Samples collected from infected plants tested positive for Cucumber mosaic virus but not Impatiens necrotic spot virus using ELISA. Mechanical inoculation and RT-PCR further confirmed the presence of Cucumber mosaic virus. Sequence analysis showed the Serbian isolate shared 100% nucleotide identity with isolates from Poland, Italy and Serbia. This represents the first report of natural Cucumber mosaic virus infection in pot marigold in Serbia.

This document describes the production of Short Chain Variable Fragments (ScFv) that specifically bind to E. coli for use in detecting E. coli in water samples. Chickens were immunized with bacterial glycoconjugates to produce antibodies. RNA from the chickens was used to generate a ScFv gene library displayed on phages. ScFvs that bound strongly to E. coli were isolated through biopanning. The purified phage was used to produce soluble ScFv in E. coli for assessment of binding specificity to E. coli and Enterobacter cloacae via dot blot analysis. EETT is a tool that identifies conserved and predicted epitopes in ebolavirus sequences. It queries the NCBI database to retrieve ebolavirus protein sequences, then compares the sequences to find conserved epitopes. EETT also queries the IEDB database to retrieve known epitopes and uses IEDB tools to predict new epitopes by analyzing ebolavirus sequences for MHC binding. The goal is to help researchers quickly gather epitope information about new ebolavirus strains to aid in the development of treatments and vaccines.

The researchers were able to obtain very high virus titers of 108 pfu/mL using the best conditions for cell propagation and viral infection in serum-free medium. The use of amoeba as “natural” containers for pathogens was utilized to develop the first living system for the transport and storage of pathogens. The amoeba system works at ambient temperature for extended periods of time—capabilities currently not available for biological sample transport.

1. The researchers collected frog egg masses from bodies of water in Illinois to isolate water mold pathogens.
2. They obtained DNA sequences from the ITS region of fourteen water mold isolates and classified the isolates using phylogenetic analysis.
3. The researchers designed a locked nucleic acid primer/probe set for quantitative PCR (qPCR) to identify a specific Saprolegnia strain. They are currently testing the probe's ability to only amplify the desired strain.

It describes how: (1) the initial antibiotic cocktail was too toxic and had to be modified by removing an antifungal and decreasing antibiotic concentrations, (2) 16S rRNA gene V3/V4 regions were amplified and sequenced, (3) sequencing reads were processed and clustered into OTUs using QIIME, and (4) alpha and beta diversity analyses were performed to characterize microbial communities. This document describes Pipestone Veterinary Services' participation in the 2017 National Institute for Antimicrobial Availability symposium on antibiotic use. Pipestone operates mixed animal practices across 5 Midwestern states and leads research on biosecurity and pig farm filtration. The company launched its PART program in 2017 to track antibiotic use on 175 subscriber farms representing 6.5 million pigs. Preliminary analysis of antibiotic resistance data from 2001-2017 found increasing then decreasing susceptibility over time across several key bacteria. A pilot study comparing farms with high versus low antibiotic use found similar bacterial recovery but differences in resistance levels. Biotechnology allows scientists to sequence genomes, cut and insert genes into chromosomes, and control gene expression. It enables knocking out genes in model organisms to study disease and evolution. Sequencing the human genome demonstrated our similarities to other species but did not fully explain genetic disease. Genomics uses sequencing and databases to identify gene locations and functions. Sequencing bacteria and viruses provides insights into their traits, transmission, and epidemiology.
PRODUCTION OF YELLOW FEVER VIRUS IN VERO CELLS GROWN IN SERUM-FREE MEDIUM
This document summarizes research on producing yellow fever virus in Vero cells grown in serum-free medium. The researchers were able to obtain very high virus titers of 108 pfu/mL using the best conditions for cell propagation and viral infection in serum-free medium. Analysis showed no difference in antigenic properties between virus cultivated in serum-containing and serum-free medium. SDS-PAGE analysis confirmed a drastic reduction in protein content for yellow fever virus cultivated in serum-free medium. Developing a cell culture process for propagating yellow fever virus in serum-free Vero cells could be an important step towards producing a new inactivated yellow fever vaccine.
