This document evaluates the anomeric recognition of galactose-binding lectins using crosslinked hemicellulose matrices. It describes extracting xyloglucan from Tamarindus indica seeds and galactomannan from Caesalpinia pulcherrima seeds, then crosslinking them with epichlorohydrin to create affinity chromatography matrices. Four lectins - PNA, ricin, frutalin, and jacalin - were applied to the matrices and their retention profiles analyzed to determine preferential binding based on the α- or β-anomeric configuration of galactose residues present in the hemicelluloses.
