This document describes the isolation and identification of bioactive natural products from marine invertebrates. The author extracted compounds from sponge and tunicate samples using progressive solvent partitioning. Two bisabolene derivatives were identified from one sample using LC-MS, NMR, and IR spectroscopy. The author also synthesized L-isoleucic acid and coupled a bisabolene amine to naproxen isocyanate. Overall, the project aimed to discover new bioactive natural products from marine invertebrates.

Isolation of Bioactive Natural Products from Marine InvertebratesKyle Planck
This document describes the isolation of bioactive compounds from marine invertebrates. Six compounds were isolated from two different sponge samples, including Aplysamine-2 which was further characterized. Nuclear magnetic resonance spectroscopy and liquid chromatography-mass spectrometry were used to determine the structures of the compounds, which were identified by comparing the data to literature, including MarinLit. Future work proposed includes testing the compounds for antifungal activity and isolating additional compounds from other sponge samples.
This document summarizes research presented at a conference on marine natural products. It discusses several projects investigating the chemical ecology of seaweed surfaces, including the antibiotic and repellent activities of bacteria associated with seaweeds and the identification of fatty acids from Halidrys siliquosa that have antimicrobial properties. It also describes work on a biodiscovery program screening seaweed extracts for bioactive compounds, noting several extracts with anti-fouling or antibiotic activities.
Marine organisms are a rich source of novel bioactive compounds and drugs. In recent years, many metabolites with potent pharmacological properties have been discovered from marine sources. One example is Ziconotide, derived from cone snail venom, which is a non-opioid analgesic approved for severe chronic pain. It works by blocking N-type calcium channels. Marine organisms like sponges, tunicates, and mollusks continue to be explored for new drug candidates, particularly for cancer, infection, and inflammation. Challenges remain in cultivating organisms and addressing toxicity for developing marine drugs.
The drugs which are obtained from marine organisms are know as marine drugs. these marine drugs are used since ancient times. chines and japanes are very famous to use these resources. And interstingly, innumarable products derived from the marine organisms in several 'crude forms' have been widely used across the globe by the traditional practitioners for thousands of years.

This document compares the properties of five natural acetogenins from the Annonaceae plant family as inhibitors of mitochondrial Complex I, the enzyme NADH dehydrogenase, and compares them to the classical inhibitors rotenone and piericidin. Two of the acetogenins, rolliniastatin-1 and rolliniastatin-2, are more potent inhibitors of Complex I than piericidin in terms of inhibitory constant and protein dependence. Rolliniastatin-2 interacts with Complex I in a way that is mutually exclusive with piericidin but not rotenone, making it the first known potent Complex I inhibitor with a binding site distinct from rotenone.

1) The document describes the total synthesis of the natural product 10-hydroxyasimicin, which contains a bis-THF motif in its central core.
2) A key step was the development of a template approach to link two alkenol building blocks and induce stereocontrol during ring-closing metathesis to generate the bis-THF framework.
3) The synthesis involved coupling the building blocks, ring-closing metathesis, asymmetric dihydroxylation, macrocycle opening and elaboration to install side chains, and final coupling and deprotection to yield the target natural product.
This review summarizes knowledge about newly isolated acetogenins from 1998-2004. Over 176 new acetogenins were isolated in this period, bringing the total number isolated to 417. The review describes the extraction, isolation, purification and structural elucidation of these compounds. It categorizes the acetogenins into different groups based on their structures and discusses the biological activities and mechanisms of action of these interesting natural products.
ACG-185-Historic Perspective on Acetogenins-Wu-TW-2010Ben Rockefeller
This document summarizes the history of research on acetogenins from the Annonaceae plant family. It discusses the initial isolation of uvaricin in 1982 which sparked interest in these compounds. Research progressed in three stages from the early 1980s to 2000, focusing on isolation, identification, and determining stereochemistry of various acetogenins. Key accomplishments included classifying compounds by structure and developing NMR methods to determine stereochemistry. Recent studies examined mechanisms of anticancer and other bioactivities. The purpose of this review is to provide an overview of acetogenin research and help clarify their modes of action to guide further development.
ACG-008-Risk-Seeds of annona squamosa-FR-2011Ben Rockefeller
This chapter discusses acetogenins from the seeds of the custard apple (Annona squamosa L.) and their health effects. The custard apple is a tropical tree cultivated for its sweet fruit. Its seeds contain high amounts of annonaceous acetogenins (ACGs), which are potent inhibitors of mitochondrial complex 1. Recent research has found ACGs show antitumor and pesticidal properties. While ACGs have potential health benefits, the seeds also present toxicity concerns that limit their use.
The document summarizes research on herbal medicines in Taiwan, including WYC-01 and WYC-02. It discusses Taiwan integrating traditional Chinese medicine into its national healthcare system, and the World Health Organization's support for traditional medicine development. It then summarizes the characteristics, mechanisms, toxicity tests, and anti-cancer effects of the herbal compounds WYC-01 and WYC-02, which were found to inhibit tumor growth with low toxicity.
This is a lecture by Dr. Jerry McLaughlin about his research into extracts of pawpaw plants, annonaceous acetogenins, in vitro, in vivo, mechanism of action, and toxicity in mice.
