This study examined the relationship between traumatic experiences and PTSD in HIV-positive adults in Cape Town. The study found that sexual assault and exposure to trauma in a war zone were significantly associated with PTSD. Those exposed to sexual assault had 10 times greater odds of a PTSD diagnosis, while exposure to war zone trauma led to 6.9 times greater odds. Overall, participants reported very high rates of exposure to traumatic events. The study highlights the need to address mental health in HIV care and conceptualize HIV/AIDS healthcare with a biopsychosocial model to address the high burden of trauma and mental illness.

Caregivers’ perspective on non-fatal deliberate self harm
This document summarizes a study examining caregivers' perspectives on non-fatal deliberate self-harm. Fifty patients who engaged in deliberate self-harm and were admitted to a hospital were evaluated. Caregivers of these patients were interviewed using a 15-item questionnaire to assess their attitudes. Common characteristics of the self-harm incidents were that they occurred when someone else was present and no suicide notes were left. The most common psychiatric diagnosis among patients was major depressive disorder. Most caregivers reported feeling shock, anger, and a need to overprotect the individual after the self-harm incident. Quantifying the process of empathy during crisis interventions
This study examined synchrony in voice pitch between clinicians and suicidal patients during crisis interventions to quantify the process of empathy. Analysis of audio recordings found that patient and clinician voice pitch were moderately synchronized. Synchrony correlated with higher ratings of therapeutic alliance during interventions but not risk assessments. Regulation of patient emotion by the clinician also correlated with stronger alliances, depending on the phase of treatment. The results suggest empathy and alliance are related to the mutual influence between patients and clinicians on an interpersonal, moment-to-moment level.

Stigma and Family reaction among Caregivers of Persons Living with Cancer
Cancer stigma refers to a negative or undesirable perception of a person affected by cancer. Stigma can be internal—it can affect self-perception of survivors, causing guilt, blame or shame. It can also be enacted, causing discrimination, loss of employment or income, or social isolation. It can come from misinformation, lack of awareness and deeply-engrained myth.The present study consisted of 300 caregivers of persons with cancer was selected based on simple random sampling, and with inclusion and exclusion criteria. Those patients satisfying the inclusion and exclusion criteria and attending both outpatient and inpatient services of cancer specialty hospital in KIDWAI Bangalore, Karnataka were selected randomly. The data was collected from the patients & caregivers of persons living with cancer who fulfill the inclusion/exclusion criteria were taken up for the study after their consent. Semi structured interview schedule were used to understand the stigma and family reaction. The interviews and the instruments were administered by research experts.

It surveyed 265 doctors and nurses who reported feelings of guilt, shame, and loss of confidence after making an error. Individuals with severe mental illness (SMI) are at greatly increased risk for trauma exposure and for the development of posttraumatic
stress disorder (PTSD). This study reports findings from a large, comprehensive screening of trauma and PTSD symptoms among public
mental health clients in a statewide community mental health system. In 851 individuals with SMI and probable PTSD, childhood sexual
abuse was the most commonly endorsed index trauma, followed closely by the sudden death of a loved one. Participants had typically
experienced an average of 7 types of traumatic events in their lifetime. The number of types of traumatic events experienced and Hispanic
ethnicity were significantly associated with PTSD symptom severity. Clients reported experiencing PTSD in relation to events that occurred
on average 20 years earlier, suggesting the clinical need to address trauma and loss throughout the lifespan, including their prolonged
after-effects.
Over the past two decades, a growing body of research has
shown that individuals with severe mental illness (SMI) are
at greatly increased risk for trauma exposure (see Grubaugh,
Zinzow, Paul, Egede, & Frueh, 2011, for a review). Although
national surveys indicate that more than half of people in the
general population report exposure to at least one event that

This document summarizes a study that examined whether PTSD symptoms mediate the relationship between sexual abuse and substance use in juvenile justice-involved youth. The study used data from 197 youth who completed psychological assessments. Results showed that PTSD symptoms significantly mediated the relationship between sexual abuse and both drug use and alcohol use. Specifically, higher PTSD symptoms were associated with increased substance use risk for youth who experienced sexual abuse. Gender did not significantly moderate these relationships. The findings suggest targeting PTSD symptoms through treatment may help reduce substance use problems for sexually abused youth in the juvenile justice system.

Common characteristics of the self-harm incidents were that they occurred when someone else was present and no suicide notes were left. The most common psychiatric diagnosis among patients was major depressive disorder. Most caregivers reported feeling shock, anger, and a need to overprotect the individual after the self-harm incident. A significant association was found between caregivers perceiving an unsympathetic family attitude and repetition of deliberate self-harm.

Quantifying the process of empathy during crisis interventions
This study examined synchrony in voice pitch between clinicians and suicidal patients during crisis interventions to quantify the process of empathy. Analysis of audio recordings found that patient and clinician voice pitch were moderately synchronized. Synchrony correlated with higher ratings of therapeutic alliance during interventions but not risk assessments. Regulation of patient emotion by the clinician also correlated with stronger alliances, depending on the phase of treatment. The results suggest empathy and alliance are related to the mutual influence between patients and clinicians on an interpersonal, moment-to-moment level.

Stigma and Family reaction among Caregivers of Persons Living with Cancer
Cancer stigma refers to a negative or undesirable perception of a person affected by cancer. Stigma can be internal—it can affect self-perception of survivors, causing guilt, blame or shame. It can also be enacted, causing discrimination, loss of employment or income, or social isolation. It can come from misinformation, lack of awareness and deeply-engrained myth.The present study consisted of 300 caregivers of persons with cancer was selected based on simple random sampling, and with inclusion and exclusion criteria. Those patients satisfying the inclusion and exclusion criteria and attending both outpatient and inpatient services of cancer specialty hospital in KIDWAI Bangalore, Karnataka were selected randomly. The data was collected from the patients & caregivers of persons living with cancer who fulfill the inclusion/exclusion criteria were taken up for the study after their consent. Semi structured interview schedule were used to understand the stigma and family reaction. The interviews and the instruments were administered by research experts.

Individuals with severe mental illness (SMI) are at greatly increased risk for trauma exposure and for the development of posttraumatic
stress disorder (PTSD). This study reports findings from a large, comprehensive screening of trauma and PTSD symptoms among public
mental health clients in a statewide community mental health system. In 851 individuals with SMI and probable PTSD, childhood sexual
abuse was the most commonly endorsed index trauma, followed closely by the sudden death of a loved one. Participants had typically
experienced an average of 7 types of traumatic events in their lifetime. The number of types of traumatic events experienced and Hispanic
ethnicity were significantly associated with PTSD symptom severity. Clients reported experiencing PTSD in relation to events that occurred
on average 20 years earlier, suggesting the clinical need to address trauma and loss throughout the lifespan, including their prolonged
after-effects.
Over the past two decades, a growing body of research has
shown that individuals with severe mental illness (SMI) are
at greatly increased risk for trauma exposure (see Grubaugh,
Zinzow, Paul, Egede, & Frueh, 2011, for a review). Although
national surveys indicate that more than half of people in the
general population report exposure to at least one event that

The study found that exposure to bullying was common, with over 40% of students reporting some exposure. Boys reported being bullied more frequently than girls. There was a significant association found between increased frequency of bullying exposure and higher levels of PTSD symptoms. While PTSD symptoms were higher overall for girls, the relationship between bullying exposure and PTSD symptoms was not moderated by gender. Students who were both bullies and victims reported the highest levels of PTSD symptoms. The findings support that exposure to bullying is a risk factor for the development of PTSD symptoms in students.

Bipolar disorder is a serious mental health disorder that is often first diagnosed during young adulthood or adolescence. Symptoms of the illness, however, also can appear in early childhood. Although once thought rare in children, diagnosis of bipolar disorder in children has significantly increased over the last decade (Papolos & Bronsteen, 2018). Despite the increased diagnosis of bipolar disorder in children, assessment and diagnosis remain challenging and controversial. This is, in part, because of the lack of research on this disorder in children and adolescents and the growing recognition that the disease can present differently in children from how it presents in adults (AACAP, 2019). Over the years, more attention has focused on the unique presentation of bipolar disorder in the young that has introduced new ways of looking at this disease and assessing it in children.
The importance of identifying the presence of bipolar disease at an early age is highlighted by data showing that adults in whom bipolar disease started at an early age have a more severe course of the illness compared with adult-onset disease. Early-onset disease is associated with a higher risk of suicide; severe mood lability and polarity; lower quality of life and greater functional impairment; higher rates of comorbidity; and a higher risk of substance use disorders compared with adult-onset disease (Papolos & Bronsteen, 2018). The study surveyed Rwandese children aged between 8 and 19 to measure traumatic exposures using an inventory of possible war time encounters and post-traumatic stress reactions against a checklist of symptoms of PTSD. Those who meet the assessed PTSD diagnosis model are categorized as the cases of probable PTSD. In a sample of 1547, the participants were divided into two. The respondents were exposed to horrific experiences during the war including witnessing killings, rape, sexual mutilation, and in some cases hiding under the corpse. In the two groups of respondents, the study has determined that the rate of probable PTSD was 62% and 54% in the first and second samples respectively. Some of the individuals had the rate that is as high as 100%. Besides, the study found out that the rate of probable PTSD was higher in women than in men. Still, the result indicates that the test on age gave inconsistent.

Breast cancer: Post menopausal endocrine therapy
Breast cancer in postmenopausal women with hormone receptor-positive (HR+) status is a common and complex condition that necessitates a multifaceted approach to management. HR+ breast cancer means that the cancer cells grow in response to hormones such as estrogen and progesterone. This subtype is prevalent among postmenopausal women and typically exhibits a more indolent course compared to other forms of breast cancer, which allows for a variety of treatment options.
Diagnosis and Staging
The diagnosis of HR+ breast cancer begins with clinical evaluation, imaging, and biopsy. Imaging modalities such as mammography, ultrasound, and MRI help in assessing the extent of the disease. Histopathological examination and immunohistochemical staining of the biopsy sample confirm the diagnosis and hormone receptor status by identifying the presence of estrogen receptors (ER) and progesterone receptors (PR) on the tumor cells.
Staging involves determining the size of the tumor (T), the involvement of regional lymph nodes (N), and the presence of distant metastasis (M). The American Joint Committee on Cancer (AJCC) staging system is commonly used. Accurate staging is critical as it guides treatment decisions.
Treatment Options
Endocrine Therapy
Endocrine therapy is the cornerstone of treatment for HR+ breast cancer in postmenopausal women. The primary goal is to reduce the levels of estrogen or block its effects on cancer cells. Commonly used agents include:
Selective Estrogen Receptor Modulators (SERMs): Tamoxifen is a SERM that binds to estrogen receptors, blocking estrogen from stimulating breast cancer cells. It is effective but may have side effects such as increased risk of endometrial cancer and thromboembolic events.
Aromatase Inhibitors (AIs): These drugs, including anastrozole, letrozole, and exemestane, lower estrogen levels by inhibiting the aromatase enzyme, which converts androgens to estrogen in peripheral tissues. PGx Analysis in VarSeq: A User’s Perspective
Since our release of the PGx capabilities in VarSeq, we’ve had a few months to gather some insights from various use cases. Some users approach PGx workflows by means of array genotyping or what seems to be a growing trend of adding the star allele calling to the existing NGS pipeline for whole genome data. Luckily, both approaches are supported with the VarSeq software platform. The genotyping method being used will also dictate what the scope of the tertiary analysis will be. For example, are your PGx reports a standalone pipeline or would your lab’s goal be to handle a dual-purpose workflow and report on PGx + Diagnostic findings.
The purpose of this webcast is to:
Discuss and demonstrate the approaches with array and NGS genotyping methods for star allele calling to prep for downstream analysis.
Following genotyping, explore alternative tertiary workflow concepts in VarSeq to handle PGx reporting.
Moreover, we will include insights users will need to consider when validating their PGx workflow for all possible star alleles and options you have for automating your PGx analysis for large number of samples. INTRODUCTION Study located in larger study entitled: Common Mental Health Disorders among HIV-Infected Individuals in SA: Prevalence, Predictors, & Validation of Brief Psychiatric Rating Scales High Prevalence of HIV infected individuals in SA/sub-Saharan Africa is unquestionable PLWHA suffer very high rates of mental illness, especially PTSD

Diagnosis of PTSD arises out of the experience(s) of traumatic events HIV/AIDS affects families that have experienced multi-generational histories of victimization & trauma (historical t) Currently: SA considered to be one of the most violent countries (includes sexual violence e.g. rape)

- 6. METHOD: Study Design, Sampling & Procedure Main study design: quantitative, cross sectional Convenience, non-probability sampling Tools: battery of questionnaires (demographic & screening tools for depression, PTSD & substance abuse) Structured clinical interview using MINI

- 14. DISCUSSION Demographic characteristics (mean age/>f): in keeping with national figures (despite convenience sampling) f of traumatic events overall was consistently high (increases burden of disease in already under-resourced communities)
- 15. DISCUSSION Sexual assault & exposure in war zone places PLWHA at risk for PTSD diagnosis: once again highlights nature of trauma & mental health aspects of HIV/AIDS. Supports existing lit Sex/gender differences: f (sexual assault), m (physical assault & assault with weapon)
