Abstract : The invention provides nucleic acid molecules that encode a substantially non toxic mutein of Clostridium
perfringens alpha toxin. In one embodiment nucleic acid encodes a mutein alpha toxin that comprises amino acid sequence of ID
NO:3 minus 18 consecutive amino acid wherein 1 deleted amino acid residues is Hises. In another amino acid sequence of ID
NO.3 minus at least 12 consecutive amino acid residues is Higes. The present invention provides nucleic acid molecule encoding
mutein in which 9 consecutive amino acid are deleted from sequence of SEQ ID NO: wherein .3 one of which is Hises and is
replaced by a single leucine residue. In SEQ ID NO.2 nucleotides 268-294 are deleted and are replaced by three nucleotide that
code a single leucine residue. It provides attenuated clostridium perfringens that have nucleic acid molecule that encode a
substantially non toxic mutein of Clostridium alpha toxin integrated into their chromosome. This integrated nucleic acid encodes
wild type alpha toxin. Thus attenuated Clostridium perfringens can be substantially non toxic.

A nucleic acid that encodes substantially non toxic mutein of Clostridium Perfringens wherein mutein has amino acid SEQ ID
NO:3 minus 9 consecutive acid where one of the deleted is Hises. Amino acid sequence SEQ ID NO:3 minus at least 12 & one of
the deleted is Hisss. SEQ ID NO:3 minus 18 & one of the deleted is Hisss. Nine amino acids that range from Tyrsz through Trp7o
are replaced by single Leu residue. The attenuated Clostridium is substantially non toxic. nontoxic mutein of Clostridium alpha
toxin integrated into their chromosome. This integrated nucleic acid encodes wild type alpha toxin. Vaccine for attenuated
Clostridium perfringens. The vaccine comprises of one or more pharamocologically acceptable buffer, excipient or adjuvant.
Vaccine is administered by one or more routes: oral, intramuscular, intravenous, intradermal, subcutaneous or intranasal . The
attenuated organism bacterial poplypeptide is avain and cytokine. A test kit to identify weather subject animal has been naturally
infected comprises antibody. The method comprising Contacting fluid from animal with antibody & determining whether the antibody
reacts with fluid sample. A Clostridium perfringens that is CPERF/AaToxin 365-054 having A TCC Deposit No. PTA7364.
