5. The sheep and goat reaction had the most similarity with respect to the cow reaction with
having very close bands at 132kDa, 68kDa, 45kDa, and close to 14.4kDa. The chicken reaction
had the least similarity with only having a band of 132kDa of the bovine serum albumin dimer in
the western blot and 68kDa of the bovine serum albumin monomer in the Ponceau stained
nitrocellulose. According to Figure 3 of the mammalian tree, the subfamily Caprinae that contain
the sheep and goat species are closer to the family Bovidae that contain the cow species. The

antibody had been made in rabbits against duck albumin then no if any bands would appear. The
ladder for all 3 gels was 132,000 (bovine serum albumin dimer), 68,000 (bovine serum albumin
monomer), 45,000 (ovalbumin), and, 14,400 (myoglobin). The Ponceau stain, due to its high
sensitivity and water solubility, makes it easier to detect protein bands on nitrocellulose. The
nitrocellulose membrane provides a more stable and permanent environment. The Ponceau stain
broke down the bovine serum albumin dimer into the monomer. The western blot used specific
antibodies against cow serum proteins to react with serum proteins from cow, sheep goat, horse,
donkey and chicken. The polyacrylamide gel electrophoresis broke protein complexes into
individual proteins and separated the protein samples based on size. The bovine serum albumin
was visible in all lanes of the different serum organisms. Our original results did not turn out
excepted because our nitrocellulose was partly ripped and only 3 lanes were visible. This may be
due to having more buffer concentration than DI water during the electrophoresis or having a
higher voltage setting.
Conclusion
Cow albumin can manipulate solvent concentrations, pH, and temperature levels making
it a good protein concentration standard. Horses are more related to donkeys with being in the
same family of Equidae. Goats are more related to sheep with being in the same subfamily of
Caprinae however; they are also closely similar to cows with being right under the family of
Bovidae sharing similar characteristics. The closer the species is with respect to taxonomy, the
more related characteristics they share.
