2. Introduccion
The genome has been sequenced. There are three segments—large (L), medium (M)
and small (S). Five proteins have been identified—an RNA dependent RNA polymerase
(RdRp), a glycoprotein N (Gn), a glycoprotein C (Gc), a nuclear protein (NP) and a non
structural protein (NSs).
in the early stage of infection, DBV induces the immune response of
monocytes by activating STAT1, stimulates macrophages to differentiateinto
M1 phenotypes, andleads to the production of pro-inflammatory cytokines
(such as TNF-α, IL-1β, and IL-6).
in SFTS patients, abnormal production of inflammatory mediators was most
associated with the severity of the disease
Dabie Bandavirus
Dabie bandavirus, also called SFTS virus, is a tick-
borne virus in the genus Bandavirus in the
family Phenuiviridae, order Bunyavirales., it causes
severe fever with thrombocytopenia syndrome
(SFTS)
3. Introduccion
TRIM-3
Tripartite motif proteins (TRIMs) belong to the E3
ubiquitin ligase family and contain up to 80 members
in mammals
The TRIMs family consists of three relatively conservative
domains motif (RBCC), namely, the RING domain, the B-box
domain, and the coiled-coil domain
Participates in a variety of biological processes including
differentiation, proliferation, transcription, and apoptosis in virus
infection, immunoregulation, and cell stress responses
As a member of the TRIMs family, has been revealed to provide a
post-translationalmechanismfor inflammation, apoptosis,and
proliferation
4. Evaluate how TRIM3 attenuates cytokine
storm caused by Dabie bandavirus
via promoting Toll-like receptor 3
degradation
General Objective
5. Methods
RNA extraction and reverse
transcription quantitative PCR
Western blot
Fundament:
Short snippets of synthetic DNA are used to select
a segment of the genome to be amplified, and then
multiple DNA synthesis sessions are used to
amplify it.
Why is this test useful in the document?:
It was used to measure the expression of
TRIM-3 mRNA and THP-1 cells.
Fundament:
Involves the use of gel electrophoresis to
separate the proteins in the sample
based on their size
Why is this test useful in the
document?:
They were transferred to a filter
incubated with antibodies to react with
TRIM-3, and see the expression of
different proteins.
6. Methods
ELISA Immunofluorescence
Fundament:
It is an immunoassay (tests between
antigen and antibody), and is identified by
means of an enzyme
Why is this test useful in the
document?:
In this study he is analyzing TRIM-3
(Protein) through its gene for which it is
encoded
Fundament:
It is used for the determination of
proteins, drugs and hormones by the
binding of Ac to the fluorochrome
(substances that absorb light and emit it).
Why is this test useful in the document?:
It was used to evaluate TRIM-3 within the
different organs that were analyzed.
7. Results
Figure 1: E Figure 2: B, D, I
In immunofluorescence markers are used
to identify the proteins in the regions at
the cellular level, they can be markers at
the nuclear and cytoplasmic/membrane
level, from this we can identify that the
expressed TRIM-3 protein is in less
quantity in the liver, more in the kidney,
then lung, then spleen
GAPDH is present in all
cell lines (test control). In
figure B we can see how
there is an expression
prominent of the positive controls of the overexpressed TRIM,
and not of the negative control. In figure I we see the interaction
of the molecules and proteins, based on the fact that they all
have an expression peak at 0.5 when TRIM-3 is inactive and they
all fall in their expression when time passes (1h), the one that is
most Express is TAB1, and the least express is JNK1/2
8. Results
Figure 4: D, E
In D we see a more decreased expression of the
proteins, in the presence of overexpressed TRIM-3
and in relation to time, the one that is most
expressed is TLR3 at 0.5 and the one that is least
expressed is normal TRIM-3
In E we see an increased
expression of the proteins with
respect to the presence of
overexpressed TRIM-3, being able
to see that the one that is most
expressed is TRIM-3 and the one
that is least expressed in relation
to time is the TLR-3, however, all
conserve an almost constant
expression regardless of time
9. Autor´s tittle What he said TRUE /
FALSE
Wang et al., 2019 He said that TRIM 3 has an overexpression
that can result in an inhibitory effect on the
porliferation and expression of cytokines of
synoviocytes in rheumatoid arthritis.
TRUE
Li Z. et al. (2022) He said that after infection with the Dabie
bandavirus there is usually a prominent
inflammatory response which should be
avoided as soon as possible.
TRUE
Yamada et al., 2018 Very strong viruses can trigger a stimulation
of the innate immune system response
which activates the TLR and generates a
protective inflammatory response that can
trigger SFTS
TRUE

Conclusion
1. In this study we can identify the importance of having previous
knowledge about molecular biology in order to have a vision towards
the future and in this way be constantly looking for ways to improve
the quality of life of people with different pathologies
2. From the different laboratory techniques, we can find new and
innovative therapeutic techniques and thus improve the outlook for
many diseases from the medical field.
