Editor's Notes
What does high fidelity mean? Discuss the two ways it maintains fidelity:1. rejects incorrect base pairs. 2. if incorrect base pair still remains, exonuclease site removes it
One focus of research - exonuclease site and how it operates
How to monitor exonuclease site? Cystine amino acid, to which a fluorescent dye can bind, is present on both finger domain and exonuclease site. Use laser microscope to see the parts. Difficulty in distinguishing between the two sites, even with powerful laser microscope
Perform mutagenesis that creates a version of T7 with no Cys on finger domain. Sequencing results will determine if successful (TGC codes for cystine, while GAA codes for glutamic acid

Although humans do not have a high fidelity protein such as T7, by learning how it works, we can try to replicate it. As a high fidelity protein, it can help eliminate mutations which may result in disease
More accurate, improved sequencing of DNA. Sequencing is becoming more prevalent now, and one day may be used to create treatment/medicine that targets individuals based on their genes
I would like to thank the Liberty Science Center tonight for hosting this event and the Partners in Science Program for giving me the oppurtunity to do research. I would like to thank Mr. Rosario and all the program staff. I would like to thank the Center for Integrated Proteomics Research at Rutgers University. I would like to thank my mentor, Dr. Lee and my research assistant, Matthew Putnins.
