This Slideshare covers the description about DUS characterization. parameters of DUS testing, criteria of DUS Testing in tetraploid cotton or American cotton
2. DUS TESTING
• DEFINATION-
• A test done before a protection is granted in respect to a new
variety based on article 7 of the UPOV ( International union
for the protection of new varieties of plants ) Convention.
• The variety must be ;
• Distinct
• Uniform
• Stable
• Novel

3. NEED OF DUS TESTING
1. TO GENERATE BASIC DESCRIPTION OF THE
CANDIDATE VARIETY USING RELEVANT
CHARACTERISTICS .
2. TO COMPARE THE CANDIDATE VARIETY WITH THE
REFERENCE OR EXAMPLE VARIETY.
3. TO DIFFERENTIATE THE CANDIDATE VARIETY
FROM THE VARIETIES OF COMMON KNOWLEDGE.

4. REQUIREMENTS OF DUS TESTING
• Before performing the DUS test there should be;
1. Subjects of the guidelines
2. Plant materials required for testing
3. Methods of examination
4. Analysis of distinctness, uniformity ,and stability
5. Grouping of varieties
6. Table of characteristics
7. Explanation of the characters
8. Literature and other references
9. Technical questionnaire

5. UPLAND COTTON
COMMON NAME = UPLAND COTTON
BOTANICAL NAME = Gossypium hirsutum L.
2n = 52 ( tetraploid )
FAMILY = MALVACEAE
GENOME = AADD
OFTEN CROSS POLLINATED CROP
Upland cotton is the most commonly planted agricultural
cotton and provides 90% of the worlds need .
The new world tetraploid cotton species are allopolyploids,
which originated by hybridization between old and new world
diploid species.

9. DUS TESTING
• SUBJECT-
These test guidelines shall apply to all varieties of tetraploid
cotton.
• MATERIAL REQUIRED –
The minimum quantity of the seed to be provided by the
applicant shall be 2000g in case of candidate or hybrid variety
and for parental line of hybrid 1000g is required.
The seeds submitted shall have at least 75% germination, 98%
physical purity and the highest genetic purity.
• CONDUCT OF TEST
The minimum duration of DUS test shall normally be at least two
independent similar growing season.

10. TEST PLOT DESIGN
• NUMBER OF ROWS – 12
• ROW LENGTH – 6m
• ROW TO ROW DISTANCE – 90cm
• PLANT TO PLANT DISTANCE – 60cm
• EXPECTED PLANTS/REPLICATION - 120
• NUMBER OF PLANTS/HILL – ONE
• NUMBER OF REPLICATION – 3
• Observation should not be recorded on plants in
border rows.

11. METHODS AND OBSERVATION
4/6/2022 11
1. The characteristics described in the table of characteristics (table 1) shall
be used for the testing of varieties, parental lines and hybrids for their
DUS.
2. For the analysis of Distinctness and stability, observations shall be made
on 30 plants or parts of 30 parts, which shall be equally divided among 3
replications (10 plants/replication).
3. For the assessment of Uniformity of characteristics on the plot as a whole
, a population standard of 0.5% with an acceptance probability of at least
95% shall be obtained. In case of a sample size of 300 plants , the number
of off types should not exceed 6.
4. All leaf characters shall be observed on the fourth fully expanded leaf
from the top of the main stem at 50% flowering stage.
5. All observations on the flower shall be made on the first day of flowering
and at anthesis.
12. 6. Observations on the boll shall be made at full maturity and before boll
bursting.
7. For the assessment of colour characteristics , the latest Royal Horticultural
Society colour chart shall be used.

13. GROUPING OF VARIETIES
The following characteristics are proposed to be used
for grouping of cotton varieties.
Species
Leaf : shape (characteristics 8)
Flower : petal colour (characteristic 15)
Flower : pollen colour ( characteristics 19)
Boll : shape (longitudinal section) ( characteristic 23)
Fibre : length ( characteristic 33)

15. • Type of assessment of characteristics indicated in
table 1 is as follows;
• MG : Measurement by a single observation of a
group of plants or parts of plants.
• MS : Measurement of a number of individual plants
or parts of plants.
• VG : Visual assessment by a single observation of a
group of plants or parts of plants.
• VS : Visual assessment by observation of individual
plant or parts of plants.
