PROJECT SUMMERY
Project : Conversion of Cellulose and Hemi-cellulose into Sugars & Ethanol
Duration : 3yrs (2007-2010)
Guide name : Dr. (Mrs) Suman Khowala (Sr. Principal Scientist & Head)
Division : Drug Development and Biotechnology
Funded by : New millennium Indian Technology Leadership Initiative (NMITLI), CSIR
Description
The objective of this project is to develop fermentable sugar and ethanol from cheap source, like
biomass which has high quantity of Cellulose and hemicelluloses and also the production Cellulase
and Xylanase enzymes in the submerged culture of Termitomyces clypeatus in different type of
biomass.
Role & Responsibility
 Assist Scientist to design an experiment, Experimental Planning and set up, perform the
experiment, Analyze the data, acts on result and report writing.
 Cellulase and Xylanase enzymes production in submerge culture of fungi, Termitomyces
Clypeatus using biomass pulp of Tamarind Kernel Powder (TKP).
 Experiment on enhancing the quantity of enzymes in culture by mixing other biomass like
Corn cob, wheat bran, water hyacinth powder, steam exploded bagasse with TKP in
different percentage in submerged culture.
 Extract the enzymatic culture.
 Assayed of extracted enzymes using IUPAC method.
 Ultra centrifugation.
 Gel filtration chromatography.
 Ion exchange chromatography.
 High pressure Liquid chromatography.
 Saccharification of steam exploded bagasse with Purified enzyme and assay the sugar
amount.
 Well trained in basic lab method like buffer preparation, reagent preparation, Dialysis,
Ultra centrifugation, Using the Laminar flow, Plucking, Autoclaving, Inoculating,
Aliquoting, Lyophilization, Centrifugation, pipetting etc.
 Keeping the laboratory well organized and well maintained.
