Bond Elut 96-well Plates
Bond Elut 96-well plate formats are best in class for flow performance and well-to-well
reproducibility. These specially designed plates are available with well depths of 1 mL and 2 mL
and in a large range of different sorbent chemistries.
VersaPlate
VersaPlate is a highly innovative, flexible design that lets you customize plates. Insert different
phases for sorbent screening or insert only enough tubes to match the number of samples to be
extracted for minimal waste. VersaPlate can be purchased in a pre-packed format or as loose
tubes.
Packed Formats for Automation
Bond Elut sorbents are also available in packed bed formats for automation platforms, such as
the Spark Holland Symbiosis, Gilson ASPEC and Gerstel MPS systems. Agilent's unique OMIX

Solid Phase Extraction
Solid phase extraction (SPE) is a form of digital (step-wise) chromatography
designed to extract, partition, and (or) adsorb one or more components
from a liquid phase (sample) onto a stationary phase (sorbent or resin).
Over the last twenty years, SPE has become the most powerful technique
available for the rapid and selective sample preparation (prep) prior to
analytical chromatography. SPE extends the lifetime of chromatographic
systems and improves qualitative and quantitative analysis. By switching
sample matrices from the original matrix to a simpler matrix environment,
subsequent analysis is often simplified, and the demand placed on an
analytical system is considerably lessened.

tubes, glass tubes, 96-well plates Buchner funnels, and various positive pressure cartridges. Supelco offers custom

YMC*Gel
Packing materials that YMC produces are of two
types, spherical and irregular, and both are rich
in variety. Spherical packing materials have a
higher homogeneity for packing than irregular
packing materials,so they have higher efficiency,
high reproducibility of packing and, low pressure
drop. Irregular packing materials are used for
preparative column and open column chromatography.
The smaller the particle size of both spherical and
irregular, the higher efficiency is obtained, but
the larger the pressure drops.

Packing Pore size
The size of the molecules to be analyzed will determine the packing pore size. Small molecules can diffuse in and
out of 80-120 Å pore packings(Ascentis Express, Ascentis, and Discovery HS), but peptides and proteins may not.
For tihis reason, it is recommended to use 200-300 Å pore-size packings(Discovery and Discovery Biowide) for
separations of peptides and proteins.

Column Diameter
Column diameter defines how much material one can inject onto a column. Conventional analytical HPLC uses
4.6mm internal diameter. However, smaller diameter columns (1.0 and 2.1mm) Provide increased sensitivity and
therefore are commonly used in limited sample applications and LC-MS. Larger diameter columns (10 mm and 21.2
mm) allow for high sample loading which is beneficial for sample collection.

Particle Size
For conventional analytical HPLC, the standard particle size is 5um. However, smaller particles provide higher efficiency
and resolution but at a higher backpressure on the HPLC system. The latest particle technology, Fused Core, combines
the benefits of higher resolution and lower backpressure into a single colunm format. Larger particles provide lower
backpressure making them amenable to large colunms such as those used in preparative chromatography.

Column Length
The conventional column length for analytical HPLC is 150mm or 250mm for high resolution. For fast analysis,
shorter columns(30 to 50mm) should be used.

Column Bonded Phase
A C18 phase is often utilized as a starting point for many separations. C18 will provide enough retention for most
non-polar and moderately polar molecules. Analysis of polar molecules or molecules that are highly functionalized
will often be enhanced by using a more polar stationary phase such as Phenyl, RP-Amides, F5, or Cyano.

Inside the chiral columns are chiral resolving agents Chiral Stationary

Partisil SAX (5 and 10 µm)
A strong anion exchanger based on quaternary ammonium groups (-NR+3 ). Supplied
in the H2PO4 form in methanol, Partisil 10 SAX has been widely reported in literature
and is best known for the separation of nucleotides. Stable over pH range 1.5-7.5 when
used in conjunction with a Solvecon mobile phase conditioning column. Obtains the
highest anion exchange efficiencies and resolution. Applicable to separations of nucleic
acids, organic acids and inorganic anions.

Partisil SCX (5 and 10 µm)
Based on aromatic benzene sulfonic acid groups. Supplied in the ammonium form.

Chiral Column Selection Guide - Column Applicability (type of samples)
Chiral-AGP : Extremely broad applicability.
Most likely the column with the broadest applicability of all chiral columns.
Separates all kinds of compounds : • amines (primary, secondary, tertiary and quartenary)
• acids (strong and weak)
• nonprotolytes (amides, esters, alcohols)

Chiral-CBH : More narrow applicability than CHIRAL-AGP.
Separates compounds containing one or more basic nitrogens together with one or more
hydrogen accepting or hydrogen donating groups
(alcohol, phenol, carbonyl, amide, ether, sulphoxide, ester, etc.).

Chiral-HSA : More narrow applicability than CHIRAL-AGP.
Separates preferentially weak and strong acids and non-protolytic compounds.
The columns overlap for some types of compounds; basic compounds can be separated on both CHIRALAGP and CHIRALCBH, acidic and neutral compounds can be separated on both CHIRAL-AGP and CHIRAL-HSA.
However, as CHIRAL-AGP is a column with an extremely broad applicability, this column should be chosen
first, if the analyte has not been separated on any of the columns. There are, however, some types of
compounds where one of the other columns may be the first choice:

pH 전극 사용 및 관리방법
* 준비 (PREPARATION )
1. pH 전극을 싸고 있는 보호용 캡을 벗기고, 증류수나 pH 버퍼용액을 사용해 전극을 헹군다. 보호용 캡은 전극 보관을
위해 필요하므로 잘 보관해 둔다.
2. 용액 보충이 가능한 기준전극 : 충전용 구멍이 고무마개로 덮여서 배송되는 전극의 경우, 재충전을 위해 충전구멍(fill
hole)에 붙여진 테이프를 제거하고 고무 마개를 열어 내부용액을 채워 넣으면 된다. 커버(Sleeve)로 덮여서 배송되는
전극의 경우, 이 커버를 아래로 밀어 내린 후, 충전구멍이 드러나도록 테이프를 제거한다. 그리고 전극과 같이 함께
보내진 내부용액을 충전구멍 밑부분까지 채워 넣는다. 전극을 사용하여 용액을 측정 할 때는 충전구멍의 마개는 열려져
있는 상태로 두어야 한다.

3. 구입 후 최초 사용시 혹은 장시간의 보관 후 사용하고자 할 때는 pH electrode를 pH 버퍼에다가 30분간 담궈 둔 후
사용한다. 이런 과정을 통해 기준전극의 junction부분이 측정에 적합한 젖은 상태를 유지하게 된다. 이로서 측정을 위한
전극의 사전준비는 완료되었다.

* 필요한 재료들(REQUIRED MATERAILS)
1. 미터 (METER)
일반 Glass Membrane pH Electrode는 모든 미터에 호환하여 사용이 가능하지만, Antimony pH electrode는 모든
pH(혹은 mV) Meter에서 사용할 수 있는 것은 아니다. 일반적인 pH Meter에서 Antimony pH electrode를 사용하는 중
발생하는 주된 문제점은 Antimony electrode와 일반 Glass pH electrode의 Millivolt 출력 값 차이에 기인한다. pH값을
정확하게 측정하기 위해서는 Antimony electrode의 약 -400mV 편차를 보정해주는, 영점 조정을 Meter에서 해주어야
한다. 어떤 pH Meter는 영점조정이 가능한 것도 있지만, 대부분이 그렇지 못하다. pH전극과 Meter와의 연결 및 영점
조정하는 방법에 대한 상세한 설명은 Meter의 사용자 매뉴얼을 참조하면 된다. 만일 pH 모드에서 영점조정이 가능하지
않을 경우, 보정과 측정을 위해 Millivolt Mode를 이용해야 한다.
2. 버퍼 (BUFFER)
정확한 전극 보정(표준화)을 위해서는 2개의 버퍼가 필요하고, 1개의 버퍼는 측정샘플의 pH에 가까운 버퍼용액을
사용하여야 한다. pH7.00(25℃에서) 버퍼를 사용하여 영점을 잡고, pH4.01(25℃에서)나 10.01(25℃에서) 버퍼를
사용하여 스팬(slope)조정을 한다.
3. 전극의 보정 (ELECTRODE STANDARDIZATION )
(1)깨끗한 pH 7.00 Buffer 에 전극을 넣고 저어준다. Meter의 사용설명서대로 Meter 가 pH 버퍼 값을 얻도록 조절해
준다. 만약에 이것이 안 된다면, Millivolt Mode를 이용하여 Millivolt 값을 읽어 측정하도록 한다.
(2)전극을 증류수로 깨끗하게 세척한 후 측정하고자 하는 샘플이 산성인지 염기성인지에 따라 깨끗한 4.01 Buffer 혹은
10.01 Buffer에 넣어준다. 측정값이 안정될 때까지 30초~1분 정도 교반을 하여준다. Meter의 사용 설명서에 언급된
대로 Meter가 pH 버퍼 값을 읽도록 조정하여 주거나, Millivolt Mode를 이용하여 Millivolt 값을 읽어 측정할 수도 있다.
(3)증류수로 전극을 깨끗하게 헹구어 낸 후, 측정하고자 하는 샘플에 넣고 교반하여 준다. 약30초에서 1분 정도
Meter가 읽는 값이 안정될 때까지 기다린다. 측정된 pH값이나, 측정된 Millivolt 값에서 산출한 pH 값을 기록한다.
정확한 측정을 위해, Buffer와 Sample의 온도를 상온으로 유지하여야 한다.

* 전극의 보관(ELECTRODE STORAGE)
전극이 최상의 상태를 유지하기 위해서는, 기준전극의 Junction부분이 젖어 있을 수 있도록 포화 KCl 용액과 pH4.01
Buffer를 1:1비율로 섞은 용액에 보관하는 것이 바람직하다. 다른 pH 버퍼용액이나 단기간의 경우 수돗물도
보관용액으로 사용할 수 있다. 그렇지만 증류수의 경우는 보관용액으로 일반적으로 사용하지 않는다.
전극의 장기간 보관을 위해서는 전극을 보관용액이 들어있는 보호용 Cap에 넣어두는 것이 바람직하다.
주의점 : 전극의 정확한 사용을 위해서는 6개월 이상 전극을 보관하여서는 안되기 때문에 재고 관리에 주의하여야 한다.
*  극의 유지보수 (ELECTRODE CLEANING )
전
기계적으로 완전한(외관상 파손된 부분이 없는)전극들은 일부의 경우, 아래의 방법들을 통해 정상적인 상태로 회복시킬
수 있다.
(1)염분의 제거 (Salt Deposits) : 먼저 0.1M HCl 에 약 5분간 전극을 넣어둔 후, 다시 0.1M NaOH용액에 약 5분간
전극을 넣어둔다. 이를 2~3회 반복 후 증류수로 깨끗하게 세척한다.
(2)Oil/Grease 막의 제거방법(Oil/Grease films) : 합성세제 및 일반적인 세제를 사용하여 Oil/Grease 막을 제거한 후
증류수로 세척한다.
(3)지시전극의 미세한 부분이 막혔을 때(Clogged Reference) : 희석시킨 KCl용액을 60~80도로 가열한다. 여기에
전극을 약 10분 정도 넣어둔다. 10분이 지난 후 가열하지 않은 KCl용액에서 냉각시킨다. 후 증류수로 깨끗하게
세척한다.
(4)단백질 제거방법(Protein Deposits) : 단백질 분해효소인 10%의 펩신에 0.1M의 HCl을 첨가하여, pH를 1~2로
맞추고 전극을 약 5분 정도 넣어두고 난 후 증류수로 전극을 세척한다.
(5)Antimony Electrode 일반적인 클리닝법 : 만일 전극의 표면이 이물질로 오염이 되었다면, 적절한 등급의 Abrasive
paper로 가볍게 마찰시켜 반짝이는 표면이 나타나게 되면, 그 전극의 기능은 대개 복구되어진다. 만일 Abrasive
paper가 없다면 뜨거운 물에 1:10 비율로 희석한 가정용 세탁물 표백제와 0.1~0.5% 액체 세정 액을 넣은 희석액에
흠뻑 적셔 15분 정도 강하게 휘저어 준다.그리고 Junction을 흐르는 미지근한수돗물(tap water)에 약 15초 정도 둔 후,
기준전극의 내부 충진 용액을 교체하여 준다. 전극을 적어도 10분 정도 Storage solution에 넣어둔다.

  • 19. Columns SUPELCO Supelco, A pioneer in Chromatography Supelco has a 30 year history in pioneering chromatography products which has led to worldwide recognition. HPLC HPLC Column Selection Packing Pore size The size of the molecules to be analyzed will determine the packing pore size. Small molecules can diffuse in and out of 80-120 Å pore packings(Ascentis Express, Ascentis, and Discovery HS), but peptides and proteins may not. For tihis reason, it is recommended to use 200-300 Å pore-size packings(Discovery and Discovery Biowide) for separations of peptides and proteins. Column Diameter Column diameter defines how much material one can inject onto a column. Conventional analytical HPLC uses 4.6mm internal diameter. However, smaller diameter columns (1.0 and 2.1mm) Provide increased sensitivity and therefore are commonly used in limited sample applications and LC-MS. Larger diameter columns (10 mm and 21.2 mm) allow for high sample loading which is beneficial for sample collection. Particle Size For conventional analytical HPLC, the standard particle size is 5um. However, smaller particles provide higher efficiency and resolution but at a higher backpressure on the HPLC system. The latest particle technology, Fused Core, combines the benefits of higher resolution and lower backpressure into a single colunm format. Larger particles provide lower backpressure making them amenable to large colunms such as those used in preparative chromatography. Column Length The conventional column length for analytical HPLC is 150mm or 250mm for high resolution. For fast analysis, shorter columns(30 to 50mm) should be used. Column Bonded Phase A C18 phase is often utilized as a starting point for many separations. C18 will provide enough retention for most non-polar and moderately polar molecules. Analysis of polar molecules or molecules that are highly functionalized will often be enhanced by using a more polar stationary phase such as Phenyl, RP-Amides, F5, or Cyano.   • 57. pH 전극 사용 및 관리방법 * 준비 (PREPARATION ) 1. pH 전극을 싸고 있는 보호용 캡을 벗기고, 증류수나 pH 버퍼용액을 사용해 전극을 헹군다. 보호용 캡은 전극 보관을 위해 필요하므로 잘 보관해 둔다. 2. 용액 보충이 가능한 기준전극 : 충전용 구멍이 고무마개로 덮여서 배송되는 전극의 경우, 재충전을 위해 충전구멍(fill hole)에 붙여진 테이프를 제거하고 고무 마개를 열어 내부용액을 채워 넣으면 된다. 커버(Sleeve)로 덮여서 배송되는 전극의 경우, 이 커버를 아래로 밀어 내린 후, 충전구멍이 드러나도록 테이프를 제거한다. 그리고 전극과 같이 함께 보내진 내부용액을 충전구멍 밑부분까지 채워 넣는다. 전극을 사용하여 용액을 측정 할 때는 충전구멍의 마개는 열려져 있는 상태로 두어야 한다. 3. 구입 후 최초 사용시 혹은 장시간의 보관 후 사용하고자 할 때는 pH electrode를 pH 버퍼에다가 30분간 담궈 둔 후 사용한다. 이런 과정을 통해 기준전극의 junction부분이 측정에 적합한 젖은 상태를 유지하게 된다. 이로서 측정을 위한 전극의 사전준비는 완료되었다. * 필요한 재료들(REQUIRED MATERAILS) 1. 미터 (METER) 일반 Glass Membrane pH Electrode는 모든 미터에 호환하여 사용이 가능하지만, Antimony pH electrode는 모든 pH(혹은 mV) Meter에서 사용할 수 있는 것은 아니다. 일반적인 pH Meter에서 Antimony pH electrode를 사용하는 중 발생하는 주된 문제점은 Antimony electrode와 일반 Glass pH electrode의 Millivolt 출력 값 차이에 기인한다. pH값을 정확하게 측정하기 위해서는 Antimony electrode의 약 -400mV 편차를 보정해주는, 영점 조정을 Meter에서 해주어야 한다. 어떤 pH Meter는 영점조정이 가능한 것도 있지만, 대부분이 그렇지 못하다. pH전극과 Meter와의 연결 및 영점 조정하는 방법에 대한 상세한 설명은 Meter의 사용자 매뉴얼을 참조하면 된다. 만일 pH 모드에서 영점조정이 가능하지 않을 경우, 보정과 측정을 위해 Millivolt Mode를 이용해야 한다. 2. 버퍼 (BUFFER) 정확한 전극 보정(표준화)을 위해서는 2개의 버퍼가 필요하고, 1개의 버퍼는 측정샘플의 pH에 가까운 버퍼용액을 사용하여야 한다. pH7.00(25℃에서) 버퍼를 사용하여 영점을 잡고, pH4.01(25℃에서)나 10.01(25℃에서) 버퍼를 사용하여 스팬(slope)조정을 한다. 3. 전극의 보정 (ELECTRODE STANDARDIZATION ) (1)깨끗한 pH 7.00 Buffer 에 전극을 넣고 저어준다. Meter의 사용설명서대로 Meter 가 pH 버퍼 값을 얻도록 조절해 준다. 만약에 이것이 안 된다면, Millivolt Mode를 이용하여 Millivolt 값을 읽어 측정하도록 한다. (2)전극을 증류수로 깨끗하게 세척한 후 측정하고자 하는 샘플이 산성인지 염기성인지에 따라 깨끗한 4.01 Buffer 혹은 10.01 Buffer에 넣어준다. 측정값이 안정될 때까지 30초~1분 정도 교반을 하여준다. Meter의 사용 설명서에 언급된 대로 Meter가 pH 버퍼 값을 읽도록 조정하여 주거나, Millivolt Mode를 이용하여 Millivolt 값을 읽어 측정할 수도 있다. (3)증류수로 전극을 깨끗하게 헹구어 낸 후, 측정하고자 하는 샘플에 넣고 교반하여 준다. 약30초에서 1분 정도 Meter가 읽는 값이 안정될 때까지 기다린다. 측정된 pH값이나, 측정된 Millivolt 값에서 산출한 pH 값을 기록한다. 정확한 측정을 위해, Buffer와 Sample의 온도를 상온으로 유지하여야 한다. * 전극의 보관(ELECTRODE STORAGE) 전극이 최상의 상태를 유지하기 위해서는, 기준전극의 Junction부분이 젖어 있을 수 있도록 포화 KCl 용액과 pH4.01 Buffer를 1:1비율로 섞은 용액에 보관하는 것이 바람직하다. 다른 pH 버퍼용액이나 단기간의 경우 수돗물도 보관용액으로 사용할 수 있다. 그렇지만 증류수의 경우는 보관용액으로 일반적으로 사용하지 않는다. 전극의 장기간 보관을 위해서는 전극을 보관용액이 들어있는 보호용 Cap에 넣어두는 것이 바람직하다. 주의점 : 전극의 정확한 사용을 위해서는 6개월 이상 전극을 보관하여서는 안되기 때문에 재고 관리에 주의하여야 한다. *  극의 유지보수 (ELECTRODE CLEANING ) 전 기계적으로 완전한(외관상 파손된 부분이 없는)전극들은 일부의 경우, 아래의 방법들을 통해 정상적인 상태로 회복시킬 수 있다. (1)염분의 제거 (Salt Deposits) : 먼저 0.1M HCl 에 약 5분간 전극을 넣어둔 후, 다시 0.1M NaOH용액에 약 5분간 전극을 넣어둔다. 이를 2~3회 반복 후 증류수로 깨끗하게 세척한다. (2)Oil/Grease 막의 제거방법(Oil/Grease films) : 합성세제 및 일반적인 세제를 사용하여 Oil/Grease 막을 제거한 후 증류수로 세척한다. (3)지시전극의 미세한 부분이 막혔을 때(Clogged Reference) : 희석시킨 KCl용액을 60~80도로 가열한다. 여기에 전극을 약 10분 정도 넣어둔다. 10분이 지난 후 가열하지 않은 KCl용액에서 냉각시킨다. 후 증류수로 깨끗하게 세척한다. (4)단백질 제거방법(Protein Deposits) : 단백질 분해효소인 10%의 펩신에 0.1M의 HCl을 첨가하여, pH를 1~2로 맞추고 전극을 약 5분 정도 넣어두고 난 후 증류수로 전극을 세척한다. (5)Antimony Electrode 일반적인 클리닝법 : 만일 전극의 표면이 이물질로 오염이 되었다면, 적절한 등급의 Abrasive paper로 가볍게 마찰시켜 반짝이는 표면이 나타나게 되면, 그 전극의 기능은 대개 복구되어진다. 만일 Abrasive paper가 없다면 뜨거운 물에 1:10 비율로 희석한 가정용 세탁물 표백제와 0.1~0.5% 액체 세정 액을 넣은 희석액에 흠뻑 적셔 15분 정도 강하게 휘저어 준다.그리고 Junction을 흐르는 미지근한수돗물(tap water)에 약 15초 정도 둔 후, 기준전극의 내부 충진 용액을 교체하여 준다. 전극을 적어도 10분 정도 Storage solution에 넣어둔다. 56 www.yaletech.kr
