Tissue processing
Rats were rapidly anesthetized with
isoflurane anesthetic (Vedco, St. Joseph,
MO) and then perfused intracardially
with 0.1 M PBS followed by 4%
paraformaldehyde (Electron Microscopy
Sciences, Hatfield, PA) in 0.1 M
phosphate buffer (pH 7.4). Immediately
after perfusion, brains were collected,
postfixed in the same fixative for 2 h,
cryoprotected in 0.1 M phosphate
containing 25% sucrose overnight at
Thirty-micrometer coronal cryostat
sections of the hindbrain were collected
for immunostaining.
BSA Blocking
Sections were incubated in 0.1% sodium
borohydride (NaBH4, w/v) in PBS),
rinsed, and incubated in 10% NHS w/
0.5% Triton X-100 for one hour to block
nonspecific staining. .
Primary antibody incubation
Sections were then incubated in rabbit
phospho-synapsin I (S605) antibody in
10% NHS with 0.5% Triton X-100
overnight at RT.
Secondary antibody incubation
Incubated in Alexa Fluor 555-
goat anti-rabbit (1:1000, Invitrogen) for
three hours in shielded vials..
Visualization
Stained sections were mounted on
and cover-slipped with ProLong Gold
(Invitrogen) prior to microscope
examination.
