In a randomized controlled trial, estriol (E3) significantly reduced hot flashes in postmenopausal women when taken daily. E3 also decreased urinary urgency and leakage as well as the frequency of urinary tract infections. Additional benefits included restoring normal vaginal flora and improving vaginal atrophy without adverse endometrial effects. E3 was also found to increase bone density without negatively impacting lipids, liver function, breast tissue, or blood pressure. Interestingly, E3 treatment improved symptoms of multiple sclerosis by reducing cerebral lesions, while lesions increased when treatment was stopped and decreased when restarted. E3's proposed mechanisms of action in neurological conditions include decreasing pro-inflammatory cytokines and increasing antibodies to autoantigens
