This document discusses the application of viral metagenomics and reverse genetics in the study of coronaviruses (CoVs), highlighting their significance in tracing zoonotic origins, understanding viral diversity, and developing therapeutics. It details methods for manipulating the genetic structure of CoVs to create infectious clones and assess their pathogenicity, as well as the challenges related to their study. The insights gained from these studies inform both current research in coronavirus emergence and the development of strategies to mitigate future viral threats.
