4. Lethal Dose (LD50)
+ It is a dose/ amount of any chemical/virus suspension
that can kill 50% of the test population in a specific
time period.
5. Embryo infectious dose (EID50)
+ It is a dose or amount of virus suspension that
can infect 5 0 % of the test population in a
specific time period.
6. Tissue culture infectious dose (TCID50)
+ It is the dose of virus which can cause cytopathic
effect (CPE) in 50% of tissue culture cells in specific
time period.
7. Procedure
+ 10-fold serial dilution should be performed( 9ml PBS
suspension, 1ml viral sample)
+ Inoculation of dilutions into the cell.
+ After five days observe cytopathic effect (CPE).
+ Calculate the dose.
8. Methods for cultivation of virus
+ Viruses can be cultivated within suitable hosts,
such as a living cell. Generally three methods
are employed for the virus cultivation.

9. An embryonated egg offers various site of inoculation:
- Allantoic cavity
- Amniotic cavity
- Chorioallantioc membrane
- Yolk sac
