Phage display is one of the most powerful and widely used laboratory technique for the study of protein-protein, protein-peptide and protein-DNA interactions. This technology is mainly based on displaying the interest protein (peptides, antibodies, scaffolds or others) on the surface of employing phage and then be used to interrogate the constructed libraries containing millions or even billions of displayed phages. Theoretically, phage display is an exogenous gene expression method which the gene encoding the interest protein is inserted into bacteriophage coat protein gene then displaying the interest protein on the phage surfaces, resulting in a connection between genotype and phenotype.
Creative Biolabs has long-term devoted to the development and application of phage display technology. With years of experience, our scientists have developed several phage display based platforms and tailored hundreds of particular libraries and thousands of specific antibody products to boost our global customers’ research and project goals. Phage Display Screening for Single Domain Antibody (sdAb)ShawVivian
Phage display screening(https://www.creative-biolabs.com/sdab/phage-display-screening-for-single-domain-antibody-sdab.htm) is a well-established technique to identify antibody fragments and other binding molecules against any targets of interest. As each clone in phage display libraries represents a noncombinatorial functional domain of a naturally circulating antibody, phage display screening is a powerful method for sdAb discovery. Here, Creative Biolabs describes the phage display screening for sdAb generation.

Oncolytic viruses encoding reporter genes utilized for in vivo molecular imaging are useful to locate the distribution of oncolytic viruses in pre-clinical tests. Optical detection methods mainly include green fluorescent protein (GFP), enhanced GFP (eGFP), discosoma red fluorescent protein (DsRed), and bioluminescence imaging (BLI), which utilizes luciferases. Reporter-encoding oncolytic viruses, including vaccinia virus, adenovirus, herpes simplex virus and vesicular stomatitis virus, allow accurate tracking of gene expression, tumor metastases, viral infection as well as assessment of gene therapy.

Vaccinia virus can accommodate more than 30 kb of foreign DNA. Foreign genes can be stably integrated into the viral genome, resulting in efficient and long-term gene expression. Oncolytic viruses are a class of antitumor agents that selectively kill tumor cells without affecting normal cells. Vaccinia virus (VACV) is a large, enveloped virus that is considered as the most potential live biotherapeutic agent because of its strong oncolytic efficacy and potent antigen presentation capability that can combine well with its natural oncolytic activities for cancer immunotherapy. Many types of modified vaccinia virus have been used for in vitro and in vivo studies, as well as clinical trials.https://www.creative-biolabs.com/oncolytic-virus/category-pre-made-oncolytic-vaccinia-virus-291.htm
Partial deletion of the HSV gene results in superior packaging capacity of >30 kb foreign DNA with low toxicity as an expression vector. Multiple modified purified oncolytic herpes simplex virus (oHSV) products can avoid evading the host immune response and reduce toxicity by gene knock-out, such as ICP0, ICP4, ICP22, ICP27 or ICP47.https://www.creative-biolabs.com/oncolytic-virus/category-pre-made-oncolytic-herpes-simplex-virus-290.htm
Oncolytic viruses are using for the treatment of cancer due to the specific antitumor activity in tumor cells. Oncolytic viruses have the potential to powerfully and selectively kill cancer cells and have shown impressive efficacy in preclinical and clinical settings. However, their potential can be restricted by inefficient delivery into the complex tumor environment. Thus, the efficient delivery of oncolytic viruses remains a significant challenge in the field of oncology, limiting their therapeutic effect. https://www.creative-biolabs.com/oncolytic-virus/approaches-to-delivery-of-oncolytic-viruses.htm
Numerous viruses are being developed pre-clinically and clinically. An investigation of all registered clinical trials in 2017 demonstrates 78 interventional trials regarding OVs. This ability for near-universal therapeutic impact in cancer makes OVs a popular therapeutic tool. Today, both preclinical and early-stage clinical trials are intensively investigating the approach to improve oncolytic virotherapy.

1. Yeast display (or yeast surface display) is a particularly
powerful protein engineering technique that uses the
expression of recombinant proteins incorporated into the
cell wall of yeast for isolating and engineering antibodies
and has been applied to isolating recombinant antibodies
with binding specificity to a variety of proteins, peptides,
small molecules and T cell receptors. Yeast is superior to
phage for human antibody display in which the eukaryotic
expression environment of yeast cells is more suitable for
human protein folding, modification and translocation for
display. Therefore, the yeast displayed antibodies are
conformationally and functionally closer to human native
counterparts.
CREATIVE BIOLABS
Creative Biolabs has developed and commercialized
a proprietary yeast display platform based on our
unique Ultraff™ yeast display technology, which
permits a wide range of innovative yeast display
services, including library construction and
screening, antibody affinity maturation, production of
specific monoclonal antibodies etc. Ultraff™ yeast
display technology enables the production of highly
efficient libraries of high diversity and is now very
efficient for high affinity antibody discovery and
antibody affinity maturation.
YEAST DISPLAY
SERVICE
Features
Yeast Display Technology
Similar to phage display, yeast display provides a direct connection between genotype and
phenotype. A plasmid containing the gene of interest is contained within yeast cells and the
encoded antibody is expressed on the cell surface;
The display level of each yeast cell is variable with each cell displaying 104
to 105
copies of
the scFv;
