1. DICLOFENAC
GENERIC NAME : DICLOFENAC
ROUTE : oral, intramuscular, rectalroute, topical gel, ophthalmic preparation( eye drops)
DOSE : 50 mg BD or 100 mg sustained release preparation OD
PHARMACOLOGICAL CLASS : NSAIDS; non-selective COX inhibitor (aryl aceticacid derivatives)
MECHANISM OF ACTION :
 An analgesic- antipyretic- anti inflammatory drug
 It has potent anti inflammatory effect
 It inhibit prostaglandin (PG) synthesis
 It gets concentrated in synovial fluid, hence preferred in arthritis
 Its anti-platelet action is short lasting
 Neutrophil chemotaxis & superoxide production at inflammatory site are reduced
THERAPEUTIC USES :
 Rheumatoid & osteoarthritis
 Bursitis
 Ankylosing spondylitis
 Toothache
 Dysmenorrhoea
 Post traumatic & post operative inflammatory conditions
 Affords quick relief of pain & wound edema.
ADVERSE EFFECTS :
 Mild epigastric pain (combined use of diclofenac with misoprostal reduces GI irritation & peptic
ulcer)
 Nausea,vomiting, headache,dizziness, rashes
 Gastric ulceration & bleeding
 Reversible elevation of serum amino transferases
 Rarely kidney damage
