The fate of glucose
varies with physiological
conditions, tissues, and
organisms. BIG PICTURE
• Greek word- glycos meaning sweet/sugar and lysis meaning
splitting.
• Glycolysis is the first step in the breakdown of glucose to
extract energy for cellular metabolism. Glycolysis consists of
an energy-requiring phase followed by an energy-releasing
phase.
• Glycolysis is a series of reactions that extract energy from
glucose by splitting it into two three-carbon molecules called
pyruvates.
• Sequence of reactions converting glucose/ glycogen to
pyruvate/lactate with production of ATP.
• Embden-Meyerhof pathway
Introduction
• In organisms that perform cellular respiration, glycolysis is
the first stage of this process. However, glycolysis doesn’t
require oxygen, and many anaerobic organisms—
organisms that do not use oxygen—also have this pathway.
• Glycolysis takes place in the cytosol of a cell, and it can be
broken down into three main phases:
1. energy-requiring phase,
2. splitting phase
3. energy-releasing phase,
• Reversal of glycolysis along with the alternate
arrangement at the irreversible steps will result in the
synthesis of glucose (Gluconeogenesis)
Stage 1 - Investment of ATP.
Glucose is phosphorylated. The
negative charge concentrates
glucose in the cell and glucose
becomes less stable.
Stage 2 – The 6 carbon sugar is
split to two 3-carbon fragments.
Stage 3 – Energy yielding phase.
The oxidation of the 3-carbon
fragments yields ATP
Glycolysis
Phosphoryl transfer reaction. Kinases transfer phosphate from ATP to
an acceptor. Hexokinase has a more general specificity in that it can
transfer phosphate to other sugars such as mannose.
ΔG°’= -4.0 kcal mol-1
The conversion of an aldose to a ketose.
Phosphoglucose Isomerase
ΔG°’= .40 kcal mol-1
Phosphoglucose Isomerase
The enzyme opens the ring, catalyzes the
isomerization, and promotes the closure
of the five member ring.
Phosphofructokinase-1
PFK
ΔG°’= -3.4 kcal mol-1
The 2nd investment of an ATP in glycolysis.
Bis means two phosphate groups on
two different carbon atoms. Di
means two phosphate groups linked
together on the same carbon atom.
PFK is an important allosteric
enzyme regulating the rate of
glucose catabolism and plays
a role in integrating
metabolism.
Phosphofructokinase-1
• The PFK reaction is the first unique and
irreversible step in the glycolytic
pathway.
• It is the “committed” in the pathway.
• In general, the enzyme catalyzing the
committed step in a metabolic pathway
is the most important control component
in the pathway.
Reverse aldol condensation; converts a 6
carbon atom sugar to 2 molecules, each
containing 3 carbon atoms.

ΔG°’= 5.7 kcal mol-1
All the DHAP is converted to glyceraldehyde 3-
phosphate. Although, the reaction is reversible it
is shifted to the right since glyceraldehyde 3-
phosphate is a substrate for the next reactions of
glycolysis. Thus, both 3-carbon fragments are
subsequently oxidized.
Triose phoshate isomerase
ΔG°’ = 1.8 kcal mol-1

Stage 3: The energy yielding phase.
Glyceraldehyde 3-phosphate DH
ΔG°’ = 1.5 kcal mol-1
1,3-BPG has a high
phosphoryl-transfer potential.
It is a mixed anhydride.
An aldehyde is oxidized to
carboxylic acid and inorganic
phosphate is transferred to form
acyl-phosphate. NAD+ is reduced
to NADH.
Notice, under anaerobic conditions NAD+ must be re-supplied.
Phosphoglycerate Kinase
Substrate-level phosphorylation
ΔG°’ = -4.5 kcal mol-1
ATP is produced from Pi and ADP
at the expense of carbon oxidation
from the glyceraldehyde 3-
phosphate DH reaction.
Remember: 2 molecules
of ATP are produced per
glucose.
At this point 2ATPs were invested and 2ATPs are produced.

ΔG°’ = 1.1 kcal mol-1
Enolase
ΔG°’ = .4 kcal mol-1
Dehydration reaction
PEP
•Substrate level phosphorylation is the synthesis
of ATP from ADP that is not linked to the electron
transport system.
Pyruvate Kinase
2nd example of substrate level phosphorylation.
The net yield from glycolysis is 2 ATP
ΔG°’ = -7.5 kcal mol-1
unstable Enol form ➠ more stable keto form

The Conversion of Glucose to Pyruvate
Glucose + 2 Pi + 2 ADP + 2 NAD+ →
2 pyruvate + 2 ATP + 2 NADH +2 H+
The Energy released from the anaerobic
conversion of glucose to pyruvate is
-47kcal mol-1.
Under aerobic conditions much more
chemical bond energy can be extracted from
pyruvate.
The question still remains: How is NAD+
supplied under anaerobic conditions? Or how
is redox balance maintained?
Under anaerobic conditions pyruvate is
converted to lactate. Exercising muscle is an
example.
The NAD+ that is consumed in
the glyceraldehyde 3-
phosphate reaction is
produced in the lactate DH
reaction. The redox balance
is maintained. The activities
of glyceraldehyde 3-
phosphate DH and Lactate DH
are linked metabolically.
What happens to the
lactate after a run?
The NAD+ that is consumed in the
glyceraldehyde 3-phosphate reaction is
produced in the lactate DH reaction. Thus,
redox balance is maintained.
The NADH that is produced in the
glyceraldehyde 3-phosphate reaction is
consumed in the lactate DH reaction. Thus,
redox balance is maintained.
Remember!
Glucose + 2 Pi +2 ADP → 2 lactate + 2 ATP + 2 H2O
Fructose and galactose feed into
the glycolytic pathway
In the liver,
when fructose
enters glycolysis
the PFK
reaction is
bypassed.

  • 4. • Greek word- glycos meaning sweet/sugar and lysis meaning splitting. • Glycolysis is the first step in the breakdown of glucose to extract energy for cellular metabolism. Glycolysis consists of an energy-requiring phase followed by an energy-releasing phase. • Glycolysis is a series of reactions that extract energy from glucose by splitting it into two three-carbon molecules called pyruvates. • Sequence of reactions converting glucose/ glycogen to pyruvate/lactate with production of ATP. • Embden-Meyerhof pathway Introduction
  • 5. • In organisms that perform cellular respiration, glycolysis is the first stage of this process. However, glycolysis doesn’t require oxygen, and many anaerobic organisms— organisms that do not use oxygen—also have this pathway. • Glycolysis takes place in the cytosol of a cell, and it can be broken down into three main phases: 1. energy-requiring phase, 2. splitting phase 3. energy-releasing phase, • Reversal of glycolysis along with the alternate arrangement at the irreversible steps will result in the synthesis of glucose (Gluconeogenesis)
  • 6. Stage 1 - Investment of ATP. Glucose is phosphorylated. The negative charge concentrates glucose in the cell and glucose becomes less stable. Stage 2 – The 6 carbon sugar is split to two 3-carbon fragments. Stage 3 – Energy yielding phase. The oxidation of the 3-carbon fragments yields ATP Glycolysis

  • 8. Phosphoryl transfer reaction. Kinases transfer phosphate from ATP to an acceptor. Hexokinase has a more general specificity in that it can transfer phosphate to other sugars such as mannose. ΔG°’= -4.0 kcal mol-1
  • 9. The conversion of an aldose to a ketose. Phosphoglucose Isomerase ΔG°’= .40 kcal mol-1
  • 10. Phosphoglucose Isomerase The enzyme opens the ring, catalyzes the isomerization, and promotes the closure of the five member ring.
  • 11. Phosphofructokinase-1 PFK ΔG°’= -3.4 kcal mol-1 The 2nd investment of an ATP in glycolysis. Bis means two phosphate groups on two different carbon atoms. Di means two phosphate groups linked together on the same carbon atom. PFK is an important allosteric enzyme regulating the rate of glucose catabolism and plays a role in integrating metabolism.
  • 12. Phosphofructokinase-1 • The PFK reaction is the first unique and irreversible step in the glycolytic pathway. • It is the “committed” in the pathway. • In general, the enzyme catalyzing the committed step in a metabolic pathway is the most important control component in the pathway.
  • 13. Reverse aldol condensation; converts a 6 carbon atom sugar to 2 molecules, each containing 3 carbon atoms. ALDOLASE 2nd stage ΔG°’= 5.7 kcal mol-1
  • 14. All the DHAP is converted to glyceraldehyde 3- phosphate. Although, the reaction is reversible it is shifted to the right since glyceraldehyde 3- phosphate is a substrate for the next reactions of glycolysis. Thus, both 3-carbon fragments are subsequently oxidized. Triose phoshate isomerase ΔG°’ = 1.8 kcal mol-1
  • 15. The fate of glyceraldehyde 3-phosphate Stage 3: The energy yielding phase. Glyceraldehyde 3-phosphate DH ΔG°’ = 1.5 kcal mol-1 1,3-BPG has a high phosphoryl-transfer potential. It is a mixed anhydride. An aldehyde is oxidized to carboxylic acid and inorganic phosphate is transferred to form acyl-phosphate. NAD+ is reduced to NADH. Notice, under anaerobic conditions NAD+ must be re-supplied.

Phosphoglycerate Kinase Substrate-level phosphorylation ΔG°’ = -4.5 kcal mol-1 ATP is produced from Pi and ADP at the expense of carbon oxidation from the glyceraldehyde 3- phosphate DH reaction. Remember: 2 molecules of ATP are produced per glucose. At this point 2ATPs were invested and 2ATPs are produced.

The Conversion of Glucose to Pyruvate Glucose + 2 Pi + 2 ADP + 2 NAD+ → 2 pyruvate + 2 ATP + 2 NADH +2 H+ The Energy released from the anaerobic conversion of glucose to pyruvate is -47kcal mol-1. Under aerobic conditions much more chemical bond energy can be extracted from pyruvate. The question still remains: How is NAD+ supplied under anaerobic conditions? Or how is redox balance maintained?
  • 21. Under anaerobic conditions pyruvate is converted to lactate. Exercising muscle is an example. The NAD+ that is consumed in the glyceraldehyde 3- phosphate reaction is produced in the lactate DH reaction. The redox balance is maintained. The activities of glyceraldehyde 3- phosphate DH and Lactate DH are linked metabolically. What happens to the lactate after a run?
