GENOTYPING OF ECHINOCOCCUS SPECIES ISOLATED FROM HUMAN &
ANIMALS

• Out of 50 animal samples tested, 56 % were confirmed as E. granulosis while
44%) were E. multicularis.
• Among humans, out of 15 samples 10 66% were E. granulosis and 34% were
E. multicularis.
• Out of 28 Echinococcus granulosus confirmed samples, 96% were confirmed
to be G1 (Genotype) sheep strain while the remaining 4 % were identifeid as
G6 (Genotype) camel strain.
• Similarly in case of human samples out of 10 Echinococcus granulosis
confirmed samples, 9 (90%) were confirmed as G1 (Genotype) while only 1
sample was found to b G6 (camel strain).

MOLECULAR CHARACTERIZATION OF OF DENGUE VIRUS PREVALENT IN
PAKISTAN

• All the four known serotypes of the Dengue virus were prevalent in Pakistan.
• Abundance of serotype 3 and 2 was recorded.
• Prevalence of more than one DENV serotypes indicates that future outbreaks
of dengue may cause more sever form of the disease due to increased
chances of secondary infection.
• 11.67% of patients were positive for DENV1, 48.33% for DENV2, 30% for
DENV3, 5% DENV4 and 5 % to have mixed infection in Punjab.
• In KPK 10.83% patients were positive for DENV1, 45% for DENV2, 32.5% for
DENV3 ,5% for DENV4 and 6.67% for mixed infection.
• In KPK 55% of patients confirmed visit to endemic areas of Punjab. 20% of
patients identified uncovered drains.12% patients had uncovered containers
in the premises of their houses, 11% patients viewed rain water reservoirs as
the source of dengue infection and 2% patients identified watered flowerbeds
as the cause of infection.

BOVINE BRUCELLOSIS IN CATTLE AND HUMANS IN DISTRICT PESHAWAR

• In humans overall sero-prevalence was recorded 15.84%, 14.35% and 12.37%
according to Serum Plate Agglutination Test (SPAT), Rose Bengal Plate Test
(RBPT) and indirect ELISA, respectively.
• In cattle highest incidence was found in abattoirs (22.77%) followed by animal
market (18.51%) and farms (14.11%) according to i-ELISA.
• Among various factors in individual cattle significant difference (P<0.05) was
found in sex, age groups, body condition score, parity, history of abortion and
history of retained placenta.
• High incidence was observed in butchers 20.58%, followed by veterinary
assistants 14.28%, farm workers 12.28%, abattoir workers 11.53%, farm owners
10.41% and no positive cases were found in veterinarians according i-ELISA.
• Comparative evaluation showed that RBPT was more sensitive and specific than
SPAT both in cattle and humans.

GENETIC RESISTANCE AND IMMUNE STATUS OF LOCAL AND EXOTIC
CATTLES AGAINST MYCOBACTERIUM TUBERCULOSIS AND SALMONELLA
TYPHIMURIUM
Naila Niaz and Sohail Ahmad
• A health record of past five years (2008-2012) of HF, Jersy and Achai was
evaluated available at Government CBDF Harichand, suggested that Achai was
less suffered from tuberculosis and Salmonellosis.
• All the cows depicted low antibody titer ≤ 2U/ml for tuberculosis comparable from
Strong positive control (SPC) 12U/ml and ≤ 1U/ml for salmonelosis SPC
=2.25U/ml; suggesting that the cattle were free of the disease may be due to
natural immunity.
• Genetic confirmation through PCR following candidate genes approach
suggested that Bovines may carry disease resistance gene (natural resistance
associated macrophage protein 1, NRAMP1) which accord resistance against M.
tuberculosis and S. typhimurium infection.
• The outcome of this study suggests that cattle may bear natural resistance to M.
tuberculosis and S. typhimurium infection by holding NRAMP1.

INCIDENCE OF BOVINE TUBERCULOSIS IN CATTLE, BUFFALOES AND
INFECTED HUMAN POPULATION IN DISTRICT PESHAWAR
Shah Nawaz and Muhammad Subhan Qureshi
• Sixty dairy farms were screened by taking raw milk samples from the bulk in
district Peshawar.
• Overall prevalence was found 5% and 1.6% for Mycobacterium bovis and
Mycobacterium tuberculosis respectively by PCR.
• Effect of herd size and farming system were found non-significant (p>o.o5).
• Human sputum samples were collected from patients in district TB control clinics.
• In human sputum samples M. tuberculosis (91.04%), M.bovis (5.97%) and mixed
infection (2.98%) was detected.
• In humans the body mass index (BMI) and age effected tuberculosis infection.
• Prevalence was found higher in lower BMI patient and age <20 years (P<0.05).
• The milk and sputum samples showed positive result for M.bovis which indicates
the threat of bovine tuberculosis to public health.

 Poor return to producers and poor food quality to the consumers
 Lack of formal coordination with:
• Livestock Markets and Slaughter Houses/univs
• KPCCI/SMEDA/KPBOIT
 Lack of standards for stakeholders and institutions
 The KP could not tap the livestock resoruces to alleviate poverty and
produce hygienic food for local consumption and export.

  • 32. 5/17/2015 Dairy Science Park - Blueprint 32 GENOTYPING OF ECHINOCOCCUS SPECIES ISOLATED FROM HUMAN & ANIMALS Maria Khan Panni and Ijaz Ali • Out of 50 animal samples tested, 56 % were confirmed as E. granulosis while 44%) were E. multicularis. • Among humans, out of 15 samples 10 66% were E. granulosis and 34% were E. multicularis. • Out of 28 Echinococcus granulosus confirmed samples, 96% were confirmed to be G1 (Genotype) sheep strain while the remaining 4 % were identifeid as G6 (Genotype) camel strain. • Similarly in case of human samples out of 10 Echinococcus granulosis confirmed samples, 9 (90%) were confirmed as G1 (Genotype) while only 1 sample was found to b G6 (camel strain).   • 33. 5/17/2015 Dairy Science Park - Blueprint 33 MOLECULAR CHARACTERIZATION OF OF DENGUE VIRUS PREVALENT IN PAKISTAN Rafi ur Rahman, MA Khan and Ijaz Ali • All the four known serotypes of the Dengue virus were prevalent in Pakistan. • Abundance of serotype 3 and 2 was recorded. • Prevalence of more than one DENV serotypes indicates that future outbreaks of dengue may cause more sever form of the disease due to increased chances of secondary infection. • 11.67% of patients were positive for DENV1, 48.33% for DENV2, 30% for DENV3, 5% DENV4 and 5 % to have mixed infection in Punjab. • In KPK 10.83% patients were positive for DENV1, 45% for DENV2, 32.5% for DENV3 ,5% for DENV4 and 6.67% for mixed infection. • In KPK 55% of patients confirmed visit to endemic areas of Punjab. 20% of patients identified uncovered drains.12% patients had uncovered containers in the premises of their houses, 11% patients viewed rain water reservoirs as the source of dengue infection and 2% patients identified watered flowerbeds as the cause of infection.   • 34. 5/17/2015 Dairy Science Park - Blueprint 34 BOVINE BRUCELLOSIS IN CATTLE AND HUMANS IN DISTRICT PESHAWAR Fida Muhammad Khan and Muhammad Subhan Qureshi • In humans overall sero-prevalence was recorded 15.84%, 14.35% and 12.37% according to Serum Plate Agglutination Test (SPAT), Rose Bengal Plate Test (RBPT) and indirect ELISA, respectively. • In cattle highest incidence was found in abattoirs (22.77%) followed by animal market (18.51%) and farms (14.11%) according to i-ELISA. • Among various factors in individual cattle significant difference (P<0.05) was found in sex, age groups, body condition score, parity, history of abortion and history of retained placenta. • High incidence was observed in butchers 20.58%, followed by veterinary assistants 14.28%, farm workers 12.28%, abattoir workers 11.53%, farm owners 10.41% and no positive cases were found in veterinarians according i-ELISA. • Comparative evaluation showed that RBPT was more sensitive and specific than SPAT both in cattle and humans.   • 36. 5/17/2015 Dairy Science Park - Blueprint 36 GENETIC RESISTANCE AND IMMUNE STATUS OF LOCAL AND EXOTIC CATTLES AGAINST MYCOBACTERIUM TUBERCULOSIS AND SALMONELLA TYPHIMURIUM Naila Niaz and Sohail Ahmad • A health record of past five years (2008-2012) of HF, Jersy and Achai was evaluated available at Government CBDF Harichand, suggested that Achai was less suffered from tuberculosis and Salmonellosis. • All the cows depicted low antibody titer ≤ 2U/ml for tuberculosis comparable from Strong positive control (SPC) 12U/ml and ≤ 1U/ml for salmonelosis SPC =2.25U/ml; suggesting that the cattle were free of the disease may be due to natural immunity. • Genetic confirmation through PCR following candidate genes approach suggested that Bovines may carry disease resistance gene (natural resistance associated macrophage protein 1, NRAMP1) which accord resistance against M. tuberculosis and S. typhimurium infection. • The outcome of this study suggests that cattle may bear natural resistance to M. tuberculosis and S. typhimurium infection by holding NRAMP1.   • 37. 5/17/2015 Dairy Science Park - Blueprint 37 INCIDENCE OF BOVINE TUBERCULOSIS IN CATTLE, BUFFALOES AND INFECTED HUMAN POPULATION IN DISTRICT PESHAWAR Shah Nawaz and Muhammad Subhan Qureshi • Sixty dairy farms were screened by taking raw milk samples from the bulk in district Peshawar. • Overall prevalence was found 5% and 1.6% for Mycobacterium bovis and Mycobacterium tuberculosis respectively by PCR. • Effect of herd size and farming system were found non-significant (p>o.o5). • Human sputum samples were collected from patients in district TB control clinics. • In human sputum samples M. tuberculosis (91.04%), M.bovis (5.97%) and mixed infection (2.98%) was detected. • In humans the body mass index (BMI) and age effected tuberculosis infection. • Prevalence was found higher in lower BMI patient and age <20 years (P<0.05). • The milk and sputum samples showed positive result for M.bovis which indicates the threat of bovine tuberculosis to public health.
