• Both glucosamine and its derivative N-acetyl glucosamine are amino-monosaccharides that
serve key biochemical functions on their own and as substrate precursors for the biosynthesis
of polymers such as glycosaminoglycans (e.g., hyaluronic acid) and for the production of
proteoglycans. Glucosamine has an excellent safety profile and has been shown to provide

reduced the appearance of hyperpigmentation. OBJECTIVES: To assess the effect of a combination of
niacinamide and NAG in a topical moisturizing formulation on irregular facial pigmentation, including specific
detection of changes in colour features associated with melanin. METHODS: This was a 10-week, double-blind,
vehicle-controlled, full-face, parallel-group clinical study conducted in women aged 40-60 years. After a 2-
week washout period, subjects used a daily regimen of either a morning sun protection factor (SPF) 15
sunscreen moisturizing lotion and evening moisturizing cream each containing 4% niacinamide + 2% NAG (test
formulation; n = 101) or the SPF 15 lotion and cream vehicles (vehicle control; n = 101). Product-induced
changes in apparent pigmentation were assessed by capturing digital photographic images of the women
after 0, 4, 6 and 8 weeks of product use and evaluating the images by algorithm-based computer image
analysis for coloured spot area fraction, by expert visual grading, and by chromophore-specific image
analysis based on noncontact SIAscopy for melanin spot area fraction and melanin chromophore evenness.
RESULTS: By all four measures, the niacinamide + NAG formulation regimen was significantly (P < 0.05) more
effective than the vehicle control formulation regimen in reducing the detectable area of facial spots and the
appearance of pigmentation. CONCLUSIONS: A formulation containing the combination of niacinamide +
NAG reduced the appearance of irregular pigmentation including hypermelaninization, providing an effect
beyond that achieved with SPF 15 sunscreen.

Abstract
Resveratrol, a phytoalexin antioxidant found in red grapes, has been shown to have both
chemopreventive and therapeutic effects against many diseases and disorders, including those
of the skin. Studies have shown protective effects of resveratrol against ultraviolet radiation-
mediated oxidative stress and cutaneous damages including skin cancer. Because many of the
skin conditions stem from ultraviolet radiation and oxidative stress, this antioxidant appears to
have promise and prospects against a wide range of cutaneous disorders including skin aging
and skin cancers. However, there are a few roadblocks in the way of this promising agent
regarding its translation from the bench to the bedside. This review discusses the promise and
prospects of resveratrol in the management of skin disorders and the associated challenges.

Abstract
Epidemiological, clinical and laboratory studies have implicated solar ultraviolet (UV) radiation
in various skin diseases including, premature aging of the skin and melanoma and non-
melanoma skin cancers. Chronic UV radiation exposure-induced skin diseases or skin disorders
are caused by the excessive induction of inflammation, oxidative stress and DNA damage, etc.
The use of chemopreventive agents, such as plant polyphenols, to inhibit these events in UV-
exposed skin is gaining attention. Chemoprevention refers to the use of agents that can inhibit,
reverse or retard the process of these harmful events in the UV-exposed skin. A wide variety of
polyphenols or phytochemicals, most of which are dietary supplements, have been reported to
possess substantial skin photoprotective effects. This review article summarizes the
photoprotective effects of some selected polyphenols, such as green tea polyphenols, grape
seed proanthocyanidins, resveratrol, silymarin and genistein, on UV-induced skin inflammation,
oxidative stress and DNA damage, etc., with a focus on mechanisms underlying the
photoprotective effects of these polyphenols. The laboratory studies conducted in animal
models suggest that these polyphenols have the ability to protect the skin from the adverse
effects of UV radiation, including the risk of skin cancers. It is suggested that polyphenols may
favorably supplement sunscreens protection, and may be useful for skin diseases associated
with solar UV radiation-induced inflammation, oxidative stress and DNA damage.

Abstract
The study of free radicals is particularly relevant in the context of human skin carcinogenesis and photoaging
because of their ability to induce DNA mutations and damaging lipid peroxidation byproducts. Therefore, it
is important to identify and evaluate agents with the ability to modulate intracellular free radicals.
Significant interest exists in evaluating the chemotherapeutic and anti-oxidant properties of resveratrol
(trans-3,4',5-trihydroxystilbene). Resveratrol is a phytoalexin, a naturally occurring compound derived from
the skin of grapes and other plants. Resveratrol was selected for evaluation because of demonstrated
chemopreventive and chemotherapeutic properties in a murine skin cancer model and other human cancer
models through a variety of mechanisms. However, the intracellular anti-oxidant properties of resveratrol
on free radicals in human skin cells in vitro is not well characterized. The purpose of this research is to
investigate the ability of resveratrol to modulate the hydrogen peroxide-induced upregulation of reactive
oxygen species (ROS) free radicals in normal human skin fibroblast cells in vitro. Hydrogen peroxide is a well
known generator of free radicals that occurs during endogenous and UV-induced oxidation processes in the
human skin and was used to upregulate ROS in normal human skin fibroblast cells. Using a flow cytometry-
based assay, the results demonstrate highly significant (P<0.001) dose-dependent reduction of intracellular
hydrogen peroxide-upregulated ROS by resveratrol at 0.01%, 0.001% and 0.0001% concentrations in human
skin fibroblasts in vitro.

Abstract
The polyphenolic natural product resveratrol (RV), best known for its occurrence in grape skin
and red wine, is considered a candidate drug for prevention and treatment of cardiovascular
diseases. This review aims to summarize the molecular effects of RV on endothelial cells,
which line the inner walls of blood vessels and play a key role in the development of those
diseases. We describe how RV enhances endothelial nitric oxide production, improves
endothelial redox balance and inhibits endothelial activation in response to pro-inflammatory
and metabolic insults. Furthermore, we summarize effects of RV on endothelial senescence,
apoptosis, endothelin-1 release, and endothelial progenitor cell function. As many of RV's
actions seem to be mediated by SIRT₁, different mechanistic possibilities how RV may lead to
SIRT₁ activation are discussed.

Abstract
BACKGROUND:
Photoaged skin is characterized by coarse and fine wrinkles. The mechanism of wrinkle formation appears to involve
changes to components of the dermal extracellular matrix. Topical treatment with all-trans retinoic acid (RA) can repair
photoaged dermal matrix; this is regarded as the 'gold standard' against which repair agents are judged. To date, little is
known regarding the ability of over-the-counter 'antiageing' products to repair photoaged skin.
OBJECTIVES:
We used a modified occluded patch test to ascertain whether topical applications of cosmetic 'antiageing' products are able
to repair photoaged human skin.
METHODS:
Commercially available test products [basic moisturizer, 'antiageing' cream containing different active complex levels (6%
active: lipopentapeptide, white lupin peptides, antioxidants, retinyl palmitate; 2% active: lipopentapeptide, white lupin
peptides, antioxidants)] were applied under occlusion for 12 days prior to biopsy and histological assessment in photoaged
volunteers (n=9). RA was used as a positive control.
RESULTS:
In agreement with previous studies, the patch-test study revealed that RA produced significant fibrillin-1 deposition in the
papillary dermis (P<0.01) but had little effect on procollagen I or matrix metalloproteinase-1 expression. The 6% total active
complex formulation, however, increased the deposition of fibrillin-1 and procollagen I (P<0.01, P<0.05, respectively).
CONCLUSIONS:
This study indicates that in an in vivo 12-day patch test an over-the-counter cosmetic product can induce changes in
photoaged dermal extracellular matrix, which are indicative of repair.
