This presentation provides information about parkinsonism or Parkinson disease and pathophysiology, classification, pharmacological action and side effects of Anti Parkinson drugs.
2. Parkinsonism is a extrapyramidal motor disorder
characterised by rigidity, tremor and hyperkinesia
with other secondary effects like dementia,
defective posture, mast like face and after several
years person enable to breath properly. It occurs
due to degeneration of neuron in SNPC (substantia
nigra pars compacta), dopaminergic tract lead to
deficiency of DA(Dopamine) in striatum, therefore
activity become dominant.
3. Parkinson’s disease is degeneration neuron in
SNPC and it lead to deficiency of DA in
striatum.
Imbalance between dopaminergic (inhibitory)
and cholinergic(excitatory) system.
Oxidation of DA by Mao-B and aldehyde
dehydrogenase generates hydroxyl free radical
(OH)
which damage DNA results in neuronal
degeneration.

95% of drug metabolised peripherally only 2%
of drug cross BBB(blood brain barrier).
Peripheral decarboxylation give rise to
posterior hypertension
It is given in combination with carbidopa
which reduces it’s dose to ¼th.

increase intracellular calcium by ip3
Dag pathway.
C.V.S:- It causes postural
hypotension.
C.T.Z:- It causes nausea and
vomitting.
ENDOCRINE:- It decreases
prolactin level in blood.
9. It is a peripheral decarboxylase inhibitor, they
does not cross the blood brain barrier itself and
given in combination with levadopa.
Combination increase half life in periphery and
important point is that in combination it
decreases short term side effects like postural
hypotension and increases long term side
effects like abnormal movement and
hallucination.
10. It is conducted that neuronal degeneration in
SNPC leads to deficiency of dopamine in
striatum is a major cause of parkinsonism.
Levodopa and carbidopa are given in
combination to give synergistic effect and make
a balance between dopaminergic and
cholinergic system. The use of anti Parkinson
drugs is increasing continuously to overcome
the symptoms of Parkinson disease.
