2. PRINCIPLE
The principle of separation in GLC is partition. Gas is
used as mobile phase, liquid which is coated on to a solid
support is used as stationary phase.
The mixture of components to be separated is converted
to vapor and mixed with the gaseous mobile phase.
The component which is more soluble in the stationary
phase travels slower and eluted latter.
3. The component which is less soluble in the
stationary phase travels faster and eluted out first.
No two components has the same partition co-
efficient for a fixed combination of stationary
phase, mobile phase and other conditions.
Hence the components are separated according to
their partition co- efficients.
4. APPLICATIONS
1. Examination of many volatile oils, plant acids,
alkaloids of opium, tobacco, conium and
bellodonna, the resins of cannabis , steroidal
compounds, cardiac glycosides, sugars and
amino acids.
2. Assay of starting materials and drug
substances.
5. 3. Quantitation of drug in formulations
4. Assay of impurities or solvents in the raw
material or in the drug substances
5. For steroidal analysis.
6. Hypotensive drug can be analyzed.
