This document describes a reporter system to study the nuclear localization of the yeast SUV3 helicase protein. The SUV3 protein is conserved in mitochondria and contains both a mitochondrial targeting sequence and potential nuclear localization sequence. The hypothesis is that SUV3 is dually localized to mitochondria and the nucleus. The approach uses site-directed mutagenesis to engineer a reporter plasmid containing a fragment of SUV3 fused to LacZ to monitor SUV3's nuclear localization. Future work will verify the reporter system to study SUV3 localization. The author acknowledges support from the Cancer Research Institute and guidance from Dr. Chen.
