Even if affecting only a small percentage of commercial pet foods, problems with pet food safety impact the entire pet food industry due to recalls and loss of consumer loyalty. Such experiences have reaffirmed the need for commercial pet food manufacturers to devote extensive resources documenting product quality.

Tosoh Bioscience, a provider of chromatographic solutions for the isolation and analysis of biomolecules, introduced a new Protein A chromatography resin designed for the purification of monoclonal antibodies (mAbs). It is well-suited for high capacity capturing of immunoglobulin out of high titer feedstock. TOYOPEARL AF-rProtein A HC-650F achieves 30% to 50% greater antibody adsorption than similar products. It exhibits dynamic binding capacities of greater than 70 g/L at residence times of 5 minutes and greater than 50 g/L at residence times of 2 minutes with feedstock titers from 1 g/L to more than 10 g/L.

If you have the luxury of time you may be able to save a bundle by staining using a very diluted antibody by staining overnight or even extending incubation time for one hour.

Temperature is used in reversed-phase HPLC to improve peak shape and efficiency for small MW compounds and is also beneficial in reducing analysis time, albeit at the cost of lowering selectivity. With proteins, peak shape in RPC is generally enhanced by parameters that stabilize a single denatured state. Temperature is one way that can dramatically affect the tertiary and quaternary structure of proteins, and thus a “denatured state”. As such, we investigated the impact of temperatures up to 90 ºC on RPC of several globular proteins and monoclonal antibodies (mAbs). While, as expected, retention decreased with increasing temperature, peak shape of most proteins was dramatically affected by temperature. However, when operated under optimal conditions, peak shape and resolution of antibodies on several commercial columns was superior on a 400 angstrom pore size BIOshell Fused-Core column. In general, under optimal conditions, the wide pore Fused-Core column exhibited narrower peak widths, higher peak heights, better resolution, and thus greater sensitivity.

It has been shown to exhibit hormone-like properties, causing concerns over possible health risks. Since 2008, several governments have supported on-going studies to investigate its safety and to establish safe levels of exposure. In the U.S., the Food and Drug Administration (FDA) set the safe level to 5 mg/kg (by body weight) per day, while, in Europe, the European Food Safety Authority (EFSA) recently lowered its safe level to 4 μg/kg (by body weight) per day. As a result of the health concerns over human exposure to BPA, this compound is now closely monitored in specific products, including children’s goods.

He is now a PI at the Institute of Immunology at Oslo University Hospital where he leads an effort to develop antibody-based proteomics. His technology is called MAP for microsphere affinity proteomics. He will talk about how MAP can be used to test thousands of antibodies in parallel.

31. N-Hydroxysuccinimide(NHS)
1-(3-Dimethylaminopropyl)−3- ethalcarbodimide hydrochloride(EDC)
Phosphate buffer saline(PBS)
Bovine serum albumin (BSA)
for 10 min to create hydroxyl(-OH) groups on the surface (Baranowska et al.,
2015;Kim and Herr, 2013). The surfaces were then washed with distilled water.
Blocking to reduce subsequent non-specific binding was then performed by
incubating the sensor chip surface with a blocking solution (3% BSA in the PBS,
pH7.4) for 30 mins.
The biosensor chip was then washed with PBS for 5 mins. Subsequently,
functionalized fiber was incubated with cTnI antibody with concentration of
100μg/mL for 1 h to immobilize the antibody tofiber surface. Alongside, various
concentrations of cTnI (2 fg/mL to 10 fg/mL with a step of 2 fg/mL) proteins
solutions were prepared by dilution with PBS in advance.

C1=2 mg/ml (nồng độ trong hũ) = 2000 ug/ml
=> V = m/C1 = 8 ug/ (2000 ug/ml) = 0.004 ml = 4 uL
V = 4 uL (lấy trong hũ dye mua)
=> VPBS = Vtotal - V = 4000 uL – 4 uL = 3996 uL (PBS có sẵn)
