This study aims to determine if subjects with CD25 gene mutations are able to express CD25 and which isoform is expressed. The researchers designed experiments using flow cytometry and western blotting to induce and detect CD25 expression in T cells after stimulation with PHA and IL-2. Preliminary results showed that the experimental design was suitable for inducing and detecting high CD25 expression by flow cytometry. Western blotting detected higher CD25 protein expression after T cell activation using a monoclonal mouse anti-CD25 antibody compared to a polyclonal goat antibody. Future experiments will validate these methods on primary T cells from patients and test for CD25 expression in cells with CD25 mutations.
